摘要
以月季(Rosa chinensis)品种杂3为试材,研究了培养基中激素浓度、光照条件及外植体取材时期和部位对愈伤组织诱导的影响。结果表明,MS培养基中添加2.5 mg/L 2,4-D和暗培养有利于月季愈伤组织的诱导;以小叶为外植体时愈伤组织诱导率高于以小复叶为外植体,且生长势较好。无菌苗生长2个月左右时从中部(顶生第6、7片叶)取样,生长3个月左右时从上部(顶生第3~5片叶)取样,小叶的愈伤组织诱导情况好于从其他部位取样。
The Rosa chinensis variety Za3 was used to study the effects of hormone concentration in culture medium, illumi- nation condition, collecting period and position of explant on callus induction. Results showed that MS+2.5 mg/L 2,4-D and dark culture was good for callus induction. Higher callus induction rate and better growth potential could be achieved if using leaflet other than trifoliolate leaves as explants. Leaflet collected from the central section (the sixth or seventh leaf from the peak) on two-month aseptic seedling or upper section (the third to the fifth leaf from the peak) on three-month aseptic seedling was better explant than the other parts.
