摘要
以体细胞胚为侵染受体,通过在共培养基添加抗氧化剂来研究抗氧化剂对橡胶树农杆菌侵染效率的影响。结果表明,参试的4种抗氧化剂中,GUS基因瞬时表达效率高低顺序为硝酸银>二硫苏糖醇>L-半胱氨酸>硫代硫酸钠,共培养基中添加20 mg/L硝酸银的GUS瞬时表达率最高,染色总胚数最高达96.7%,显著高于对照76.7%。因此,抗氧化剂的应用有助于提高体细胞胚的农杆菌侵染效率。
The receptor material easily being browning and necrosis significantly decreases the frequency of the infected cells during Agrobacterium-mediated transformation. In this paper, somatic embryo as starting material was used for transformation experiments and the effects of four anti.3xidants on infection efficiency were studied. The results showed that the efficiency of GUS gene instantan,eous expression from high to low was silver nitrate, dithiothreitol, L-homocysteine and sodium thiosulfate, about of 96.7% somatic embryos could be stained on cocultivation medium supplemented 20 mg/L silver nitrate medium. Therefore, application of antioxidants was helpful to improve the efficiency of Agrobacterium-mediated somatic embryo transformation in the result of A grobacterium overgrowth being suppression.
