摘要
对基因重组菌株发酵生产γ-癸内酯的能力进行研究。对实验室构建的Yarrowia lipolytica基因重组菌株TP12与野生型菌株的生长能力及生成γ-癸内酯的能力进行比较。之后对重组菌株进行单因素条件优化,并在单因素条件优化的基础上进行正交实验优化。在最优发酵条件下,经过84h的发酵,γ-癸内酯产量最高得到3.337g/L。基因重组菌株TP12产γ-癸内酯的能力较野生型有大幅度的提高,为更高产γ-癸内酯基因重组菌的构建及其条件优化提供了参考。
To study the ability of y-decalactone production by recombinant strains. At first, compared the ability to grow and produce y-decalactone between recombinant strain Yarrowia lipolytica TP12 built by my laboratory and the wild-type. Then optimized conditions by orthogonal experiments basing on single factor experiments. Based on the optimal conditions, after 84 hours' fermentaion, the yield of y-decalactone was 3.337 g/L. Compared to wild-type, the ability to produce γ-decalactone of recombinant strain Yarrowia lipolytica TP12 had a great increase, providing a reference for building more productive recombinant strain and conditions' optimizing.
