摘要
目的观察淫羊藿苷对衰老大鼠睾丸组织基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)表达的影响。方法 SD大鼠30只,随机均分为正常组、模型组、淫羊藿苷(80 mg/kg)组。大鼠颈背部皮下注射D-半乳糖(200 mg.kg-1.d-1)造成大鼠亚急性衰老模型,同时灌胃给予药物组,每天1次,连续给药8 w。取出各组动物睾丸组织,测血清睾酮含量;取睾丸组织进行病理组织学观察;RT-PCR方法检测MMP-2及MMP-9 mRNA表达,免疫组化方法检测睾丸组织MMP-2及MMP-9表达。结果与正常组比较,模型组血清睾酮水平下降;病理学检测可见生精上皮层变薄,各级生精细胞减少;MMP-2、MMP-9 mRNA与蛋白表达降低。淫羊藿苷组明显改善上述指标(P<0.01或P<0.05)。结论淫羊藿苷对D-半乳糖所致的衰老大鼠睾丸损伤有明显的保护作用,其机制可能与上调MMP-2、MMP-9水平有关。
Objective To investigate the effect of Icaririn on expression of MMP-2 and MMP-9 of testis tissue in D-galactose-induced aging rats.Methods 30 SD rats were randomly divided into three groups: normal,model and Icaririn group(80 mg/kg).The subacute aging model of rats were induced by D-galacotose(200 mg·kg-1·d-1) following s.c.administration,at the same time,Icaririn was given i.g.for 8 weeks.Testes of each group were removed from the scrotum.Serum level of testosterone was measured respectively.Testicular tissue morphology was observed by light microscopy.The expressions of MMP-2 and MMP-9 mRNA in testis tissue were examined using RT-PCR method.The expressions of MMP-2 and MMP-9 antibodies were also immunohistochemically investigated.Results Compared to the control group,the level of serum testosterone in testis was decreased in model group.The histopathological examination suggested that seminiferous tubule diameters and various grade of spermatocytes in the testis markedly shrunk.Compared with control group,the expressions of MMP-2 mRNA,MMP-2 protein and MMP-9 mRNA,MMP-9 protein were significantly increased in model group.These alterations were significantly attenuated in Icariin group(P〈0.01).Conclusions Icariin can evidently relieve testicular damage in D-galactose-induced aging rats,which might be related to improve the expressions of MMP-2 and MMP-9.
