摘要 目的探讨HOXA10基因在卵巢中颗粒细胞的表达及其与多囊卵巢综合征的关系。方法采用逆转录聚合酶链反应及免疫印迹法分别测定25例多囊卵巢综合征(PCOS)妇女和32例卵巢功能正常(Non-PCOS)妇女卵巢黄素化颗粒细胞中HOXA10mRNA和蛋白的表达水平。结果 PCOS妇女黄素化颗粒细胞中HOXA10mRNA的表达和蛋白的表达均低于Non-PCOS妇女(P<0.01,P<0.05)。结论 HOXA10基因在卵巢中有表达,PCOS妇女卵巢黄素化颗粒细胞HOXA10mRNA和蛋白表达水平明显低于Non-PCOS妇女,提示该基因可能参与了PCOS的发生及发展过程。 Objective To determine the impact of the levels of HOXA10 in ovarian granulosa cells (GCs) on the pathogenesis involved in poIycystic ovarian syndrome. Methods GCs were obtained during the oocyte retrieval from a total of 57 women: 25 women with PCOS, and 32 women with functional ovary as the control group, . The mRNA expression of HOXA10 was measured by reverse transcription polymer- ase chain reaction (RT PCR), and western blot analysis was used to detect the expression of HOXA10 protein in GCs. Results The mRNA expression of HOXA10 in GCs was significantly lower in women with PCOS than in control (P^0.01), and the protein expression of HOXA10 in GCs was also significantly lower in women with PCOS than in controls (P〈0.05). Conclusion The mRNA and protein expressions of HOXA10 in GCs were significantly lower in women with PCOS than in the control group, which sug- gests that HOXA10 may participate in the process of PCOS.
