CTGF-siRNA转染对血管平滑肌细胞MMP-2、MMP-9表达的影响 被引量:4

Effect of small interference RNA targeting connective tissue growth factor transfection on expression of MMP-2 and MMP-9 in vascular smooth muscle cells

摘要 目的通过特异性下调结缔组织生长因子(connective tissue growth factor,CTGF)的表达,观察其对大鼠血管平滑肌细胞中金属蛋白-2(MMP-2)、金属蛋白-9(MMP-9)表达的影响。方法培养大鼠血管平滑肌细胞,免疫组化方法对细胞进行鉴定,用脂质体法将以CTGF第483号编码区为起始设计的小片段干扰核糖核酸(CTGF483-siRNA)对细胞进行转染,Western blot法检测细胞中结缔组织生长因子(CTGF)、MMP-2、MMP-9的表达。结果经免疫组化鉴定,细胞特异性染色呈阳性反应。转染组的CTGF表达明显降低,MMP-2、MMP-9的表达也受到抑制(P<0.05)。结论大鼠血管平滑肌细胞中下调CTGF的表达,可以抑制VSMC中MMP-2、MMP-9的表达,延缓动脉粥样硬化的发展过程,有望成为颈动脉粥样硬化性缺血性脑血管病的预防和治疗新靶点。 Objective To observe the effect of connective tissue growth factor(CTGF) on expression of matrix metalloproteinase-2 (MMP-2) and MMP-9 in vascular endothelial cells(VSMC). Methods Primary VSMC were cultured and identified with immunohistochemical staining. CTGF483-siRNA was transfected into VSMC with liposome. Expression of CTGF, MMP-2 and MMP-9 in VSMC were detected by Western blot. Results Immunohistochemical staining showed positive reaction in VSMC which were specifically stained. The expression levels of CTGF, MMP-2 and MMP-9 were significantly lower in VSMC after transfection than before transfection(P〈0.05). Conclusion Down-regulation of CTGF expression can inhibit the expression of MMP-2 and MMP-9 in VSMC of rats, effectively, delay the development of atherosclerosis, and thus may become a new target for the prevention and treatment of carotid atherosclerotic ischemic cerebral vascular disease.
