白术对慢传输型便秘大鼠结肠黏膜NO及NOS的影响 被引量:33

摘要 目的:应用免疫组化方法检测白术对慢传输型便秘(STC)大鼠结肠黏膜NO及NOS的影响以从微观进一步阐释白术治疗STC的机制。方法:随机将成年健康SD大鼠60只分为随机分为空白对照组、模型对照组、白术低剂组(30 g生药/kg)、白术中剂组(60 g生药/kg)、白术高剂组(120 g生药/kg),每组12只,应用大黄将除空白对照组以外的大鼠制造慢传输型便秘模型。成功造模后,空白及模型对照组给予生理盐水灌胃2 ml,白术低、中、高剂组分别给予3 g生药/ml、6 g生药/ml、12 g生药/ml的白术水煎液2 ml,早晚2次,连续15天。15天中连续观察大鼠皮毛光泽、活动范围、进食量、排便情况及体重变化。末次给药后硝酸还原法检测NO含量,NOS免疫组化法染色及半定量分析。结果:模型组大鼠结肠较其他各组粗大、迂曲,肠腔内有较大块较多粒粪便存留;各组差异均有统计学意义(P<0.01)。各组大鼠在给药前体重差异均无统计学意义(P>0.05);而在处死前的各组大鼠体重差异均显出统计学意义(P<0.01);各组大鼠远端结肠黏膜NO含量差异均存在统计学意义(P<0.01)。NOS免疫组化法染色半定量分析各组差异有显著性意义(P<0.05)。结论:白术单用能通过改变STC大鼠结肠黏膜NOS的表达而降低NO的合成,以达到治疗STC目的。其中剂量为60 g的生白术能最把上述效应发挥最好。 objective: to use immunohistochemical method detects the influence of Atractylodes macrocephala koidz on the level of NO and NOS in slow transmission constipation rats colonic mucosa and further illustrates the mechanism of STC large-headed atractylodes treatment.Methods: random will adult health SD rats 60 only divided into randomly divided into blank control,model control group,atractylodes low agent group(30 g crude/kg),atractylodes macrocephala koidz agent group(60 g crude/kg),atractylodes high agent group(120 g crude/kg),each group of 12 only,the application of rhubarb except outside of the control group will be blank rat manufacturing slow transmission constipation model.The success made,blank and model control group give saline irrigation stomach 2 ml,white atractylodes low,medium and high agent groups were given crude 3 g/ml,6 g/ml crude,12 g/ml of crude large-headed atractylodes decocting 2 ml,morning and evening 2 times,for 15 days.15 days of continuous observation rats fur burnish,activities range,food intake,defecate and weight change.The end time after the treatment method to detect NO content of nitrate reductase,NOS immunohistochemical staining method and half quantitative analysis.Results: the model rats than other bulky,colon each author,there were a large presence of more waste remained;The differences are statistically significant(P0.05);And in the group to be put to death before rats are show statistically significant weight difference(P〈0.01);The rat distal colonic mucosa NO content difference there are statistically significant(P〈0.01).NOS immunohistochemical method dyeing half quantitative analysis each significant difference(P〈0.05).Conclusion: white atractylodes can use by changing the STC rats colonic mucosa of expression and reduce NO NOS synthesis,in order to achieve the purpose STC treatment.One dose for 60 g life the most large-headed atractylodes can play the best effect.
