摘要
目的观察电针丰隆穴对高脂血症大鼠血脂及ATP结合盒转运子A1(ATP binding cassette transporterA1,ABCA1)、过氧化体增殖物激活型受体α(peroxisome proliferator-activated receptor,PPARα)mRNA及蛋白表达的影响。方法选取32只SPF级SD大鼠制备高脂血症模型,造模成功后,随机分为模型组、治疗组、对照组及治疗对照组,每组8只,另选取8只大鼠作为正常对照组。治疗组及治疗对照组大鼠进行电针丰隆穴治疗,每天1次,连续治疗30天。模型组、治疗组大鼠干预期间予高脂饲料,正常对照组、对照组及治疗对照组予普通饲料。检测各组总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipoprotein cho-lesterol,HDL-C)含量,RT-PCR及Western blot检测各组大鼠肝脏组织ABCA1、PPARαmRNA及蛋白表达的变化。结果与正常组比较,模型组大鼠TC、LDL-C水平升高,肝脏组织ABCA1、PPARαmRNA及蛋白表达降低,差异均有统计学意义(P<0.01,P<0.05),而TG和HDL-C变化不明显(P>0.05)。与模型组比较,各干预组大鼠TC、LDL-C水平降低,差异均有统计学意义(P<0.01);治疗组及治疗对照组大鼠ABCA1、PPARαmRNA及蛋白表达升高,对照组PPARαmRNA及蛋白表达升高,差异均有统计学意义(P<0.01,P<0.05);与对照组比较,治疗对照组大鼠TC、LDL-C水平降低,ABCA1、PPARαmRNA及蛋白表达升高,差异均有统计学意义(P<0.01),血浆TG和HDL-C变化不明显(P>0.05)。结论电针丰隆穴能够明显下调高脂血症大鼠血脂中TC、LDL-C水平,上调肝脏ABCA1、PPARα的基因表达,从而促进由其介导的胆固醇逆转运,对高脂血症具有一定治疗的作用。
Objective To explore the effects of electroacupuncture (EA) at Fenglong (ST40) on blood lipids, mRNA and protein expression of ATP binding cassette transporter A1 (ABCA1) and peroxisome prolifera- tor-activated receptor a (PPARcc) in hyperlipidemia rats. Methods Thirty-two SD rats of SPF grade were select- ed to prepare the hyperlipidemia model. After successful modeling, they were randomly divided into the model group, the treatment group, the control group, and the treatment control group, 8 in each group. Another 8 rats were selected as the normal control group. Rats in the treatment group and the treatment control group received EA at Fenglong (ST40), once daily for 30 successive days. Rats in the model group and the treatment group were administered with high fat forage, while rats in the normal control group, the control group, and the treat- ment control group were administered with common forage. The contents of the total cholesterol (TC), triglycer- ide (TG), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were detected in every group. The mRNA and protein expressions of ABCA1 and PPARcc were detected using RT-PCR and Western blot. Results The levels of TC and LDL-C increased, and the mRNA and protein expressions of ABCA1 and PPARcx decreased in the model group when compared with the normal group, showing statisticaldifference (P〈0.01, P〈0.05). But there was no obvious change in TG or HDL-C (P〉0.05). Compared with the model group, the levels of TC and LDL-C decreased in each intervention group with statistical difference ( P 〈 0. 01 ). The mRNA and protein expression of ABCA1 and PPARa in the treatment group and the treatment control group increased. The mRNA and protein expressions of PPARcx increased in the control group with statistical difference ( P 〈0.01, P 〈0. 05). Compared with the control group, the levels of TC and LDL-C decreased, the mRNA and protein expressions of ABCA1 and PPAR~ increased in the treatment control group with statistical difference ( P 〈0.01 ). But there was no obvious change in TG or H DL-C ( P 〉0.05). Conclusions EA at ST40 could obviously down-regulate the levels of TC and LDL-C in hyperlipidemia rats, up-regulate the expressions of ABCA1 and PPARa, thus promoting counter transport of cholesterol. It had some therapeutic effects on hyperlipi- demia.
