实验性胃溃疡大鼠自愈期间胰腺组织三叶因子3水平的变化 被引量:4

Changes of TFF3 expression in pancreatic tissue during self healing of rats with experimental gastric ulcer

摘要 目的:探讨实验性胃溃疡(GU)期间大鼠胰腺组织中三叶因子3(TFF3)的表达及变化。方法:SD大鼠共30只,随机分为GU1、GU4、GU10、GU23和正常组5组,每组6只。GU1、GU4、GU10和GU23组通过胃前壁黏膜下注射冰乙酸制备GU模型,并分别于造模后1、4、10和23d取材。正常组不做任何处理。应用免疫组化SP法检测胰腺组织TFF3蛋白和胰岛素的表达,RT-PCR法检测胰腺TFF3mRNA,ELISA法检测血清TFF3水平。结果:5组大鼠胰腺组织TFF3蛋白和胰岛素的表达水平、胰腺TFF3mRNA和血清TFF3水平差异有统计学意义(F分别为9.276、12.362,166.600,21.316,P均<0.001)。造模4、10d后胰腺组织TFF3蛋白表达水平升高,23d时仍处于较高水平(P<0.05);胰岛素变化趋势与TFF3一致;造模4d后,胰腺组织中TFF3与胰岛素的表达存在正相关(r=0.826,P<0.05)。胰腺TFF3mRNA和血清TFF3水平在造模4d后达最高,10d时略有下降,23d时进一步降低但仍高于正常组(P<0.05)。结论:胰岛TFF3与胰岛素共同释放入血,参与GU自愈期间胃黏膜的修复。 Aim : To study the changes of trefoil factor 3 (TFF3) expression during the self healing of gastric ulcer (GU). Methods: A total of 30 SD rats were allocated into 5 groups : the control group and GU1, GU4, GUIO, GU23 groups, and 6 in each group. The rats in GU1, GU4,GUIO, GU23 groups were made the GU model induced by injecting acetic acid into submucosa of gastric anterior wall,and the following indexes were detected at 1,4,10,and 23 days after the injection, respectively. The expressions of TFF3 and insulin in the pancreas were detected by immunohistochemistry SP method,TFF3 mRNA was detected by RT-PCR, serum content of TFF3 was detected by ELISA. Rosults:There were significant differ- ences in the expressions of TFF3 and insulin,TFF3 mRNA in the pancreas,and serum content of TFF3 among the 5 groups ( F was 9. 276,12. 362,166. 600, and 21. 316, respectively, P 〈 0. 001 ). The expressions of TFF3 in pancreas of GU4, GU10 groups were high,in those of GU23 group decreased,but still higher than the control group( P 〈 0.05 ). The changes of insulin was similar and positively related to TFF3 ( r = 0. 826,P 〈 0. 05 ) 4 d after the injection. The TFF3 mRNA expres- sion increased from day 1, arrived the peak on day 4, decreased a little on day 10, and the level on day 23 was similar to day 1, but still higher than control group ( P 〈 0.05 ). Conclusion :TFF3 may participate in the self healing of GU by increasing transcriptional level and releasing to blood together with insulin.
