GRP78在正常与肥胖女性患者网膜脂肪组织中表达水平的比较研究 被引量:1

摘要 目的探讨葡萄糖调节蛋白78(GRP78)在肥胖及肥胖相关疾病中的作用。方法将30例40~60岁女性患者分为3组:正常对照组(NC组,BMI<23kg/m2),超重组(OW组,BMI<28kg/m2)和肥胖组(OB组,BMI>28kg/m2),每组10例。在其腹部择期手术时取其网膜脂肪组织,应用RT-PCR和Western-blot检测上述三组患者脂肪组织中GRP78的表达水平。结果 OB组GRP78mRNA与蛋白水平(GRP78/β-actin)显著高于对照组(P<0.05)。GRP78/β-actin与BMI呈正相关(r≥0.75,P<0.05)。结论 GRP78表达量与BMI呈正相关(GRP78蛋白及mRNA水平OB组>OW组>NC组)。GRP78可作为预测肥胖及T2DM和心血管疾病危险因素的指标之一。 Objective To investigate the effect of GRP78 on the pathogenesis of obesity and obesity-related diseases. Methods 30 female patients aged between 40 and 60 years were divided into three groups(n= 10, each) : normal control group (NC, BMI〈23 kg/m2 ), overweight (OW, BMI〈28 kg/m2) and obesity group (O13, BMI〈28 kg/m2 ). Omental fat from 30 patients undergoing selective surgery was taken. Western blot and RT-PCR were applied to monitor the gene expression difference in GRP78 among three groups. Results The expression of GRP78 in omental fat were higher in obesity group than in normal control group (P〈0. 05), the expressions of GRP78 in three groups were positively correlated with BMI. Conclusions The expression of GRP78 in omental fat is significantly correlated with BMI, and GRP78 is a better predictor of risk factors for type 2 diabetes and cardiovascular disease.
