摘要
目的观察真核质粒在机体细胞内的表达时效,为临床基因治疗的给药间隔提供理论依据。方法取PCDNA3.1(+)-EGFP,pEGFP-C3,pIRES-GFP 3种绿色荧光真核质粒,利用脂质体2000转染机体细胞,观察绿色荧光蛋白在细胞的表达时间推断质粒在机体内的表达时效。结果 3组绿色荧光均在12 h开始表达,48 h达高峰,持续35 d后衰减。结论在基因治疗中真核质粒连接目的基因能在机体细胞内稳定表达35 d,基因给药间隔可选定为30 d。
Objective To observe the expressive time of eucaryon plasmid in the body cell to provide theoretical basis for interval dose of clinical gene therapy. Methods The three kinds of green fluorescence eucaryon plasmid, PCDNA3. 1 ( + )-EGFP, pEGFP-C3, pIRES-GFP were added in the liposome 2000 to transfect the body ceils and observe the expressive time of the green fluorescence to decide the expressive time of plasmid in vivo. Results The green fluorescence of the three group was expressed in 12 hours, reaching peak in 48 hours and beginning to fade after 35 days. Conclusion The objective gene on the eucaryon plasmid can express 35 days stably in the body cells, so the gene interval dose is determined as 30 days.
