地卓西平马来酸盐动物行为模型研究 被引量:2

摘要 目的 制作灵长类动物谷氨酸N 甲基 D 天冬氨酸受体非竞争性拮抗剂地卓西平马来酸盐 (MK 80 1)功能低下行为模型 ;研究奥氮平治疗谷氨酸功能低下的疗效。方法 给CebusApella猴皮下注射MK 80 1(0 0 6mg/kg,0 0 8mg/kg ,0 10mg/kg)或 /和奥氮平 (0 0 2mg/kg ,0 0 4mg/kg ,0 0 8mg/kg) ,用自编 7分量表评定药物所致动物的行为变化。结果 注射药物 3h内动物的行为表现及量表评分 (中位数 ) :0 0 6mg/kgMK 80 1动物表现中度的运动减少 (3 2 9分 ) ,运动迟缓 (3 2 5分 ) ,有 2只动物表现为急性肌张力障碍 (0 33分 ) ;0 0 2mg/kg和 0 0 4mg/kg奥氮平无任何拮抗作用 ,0 0 8mg/kg奥氮平能明显或完全拮抗动物的上述行为。 0 0 8mg/kg和 0 10mg/kgMK 80 1诱发出动物严重的运动减少 (4 6 3分 ,4 5 0分 )、运动迟缓 (4 6 3分 ,4 33分 )和全部动物的急性肌张力障碍 (4 45分 ,4 0 8分 ) ;0 0 8mg/kg奥氮平能部分拮抗 0 10mg/kgMK 80 1的行为 (2 92分、2 92分和 2 6 7分 )。奥氮平不影响MK 80 1的流涎 ,却恶化MK 80 1所致的共济失调。结论 MK 80 1是一种有用的精神分裂症药理学模型工具药 ; s Objectives The study was to study the glutamate deficiency model in Cebus Apella and the efficacy of atypical antipsychotics olanzapine in treating the glutamate deficiency model in six Cebus Apella monkeys Methods Dizolcipine maleate (MK 801,0 06, 0 08 and 0 10 mg/kg), as the non competitive NMDA receptor antagonist, and the atypical antipsychotic, olanzapine (0 02, 0 04 and 0 08 mg/kg), were administrated by subcutaneously injection A 7 scores scale was used to evaluate the behaviors of animal before and after the drug injection Results Animals with low dose of MK 801 experienced decreased locomotion (3 29), bradyskinesia (3 25) and dystonia in two of animals Low and middle dose olanzapine had no effect on these abnormal behaviors, while high dose antagonized effectively or completely these behaviors Middle and high dose MK 801 induced severe dystonia (4 45, 4 08), decreased locomotion (4 63, 4 5) and bracyskinesia (4 63, 4 33) High dose olanzapine reversed partly these behaviors of high dose MK 801(2 67, 2 92 and 2 92) By the way, olanzapine had no any effect on salivation, while, exacerbating ataxia of MK 801 Conclusions Similar to Phencyclide, MK 801 is another useful tool of psychopharmacologic model for evaluation the effects and adverse effects of antipsychotics Olanzapine, an atypical antipsychotics, is able to antagonize the abnormal behaviors induced by glutamate deficiency, which means its potiential clinical profile in clinical practice
