外源激素配比对药用植物锁阳愈伤组织诱导的影响 被引量:2

摘要 目的筛选最佳激素组合,提高锁阳组织培养效率。方法以锁阳肉质茎维管组织作为外植体,以MS为基本培养基,基于正交实验设计研究三种外源激素配比(NAA、2,4-D和6-BA)以及蔗糖和琼脂浓度对锁阳愈伤组织诱导的影响。结果 NAA、2,4-D和6-BA三种激素对锁阳愈伤组织诱导均有显著影响,不同激素之间对出愈率的作用效应大小为2,4-D>NAA>6-BA,诱导锁阳愈伤组织最佳激素配比为MS+NAA(1.0mg/L)+2,4-D(1.5mg/L)+6-BA(2.0mg/L)。培养基中琼脂浓度和蔗糖浓度对锁阳愈伤组织诱导率有一定影响,蔗糖浓度以30g/L,琼脂浓度以4.5g/L时愈伤组织诱导率最高。结论锁阳愈伤组织诱导最适培养基为MS+NAA(1.0mg/L)+2,4-D(1.5mg/L)+6-BA(2.0mg/L)+蔗糖(30g/L)+琼脂(4.5 g/L)。 Objective To improve the efficiency of Cynomorium songaricum callus culture. Methods The ascular tissue parts of fleshy stem of C. songaricum were determined as the explants and cultured in MS culture media with different culture condition for callus induction. The effect of three plant hormone proportions (NAA, 2,4 -D and 6 -BA) on the callus induction was investi- gated with orthogonal test design, and the effect of different concentrations of sugar and agar on the callus induction was studied. Results NAA, 2,4 -D and 6 -BA concentration variation had a remarkable effect on the callus, respectively. Among the three plant hormones, the effect of callus induction rating was 2,4 - D 〉 NAA 〉 6 - BA. The best phytohormone proportion of callus in- duction was : MS + NAA (1.0mg/L) + 2,4 - D (1.5mg/L) + 6 - BA (2.0rag/L). The most suitable concentrations of sugar and agar were 30g/L and 4.5g/L respectively. Conclusion The optimized medium was MS + NAA (1.0rag/L) + 2,4 - D (1.5mg/L) + 6 - BA (2.0mg/L) + sugar (30g/L) + agar (4.5 g/L). The results provided a basic foundation for producing C. songaricum callus.
