摘要
采用水浴回流和闪式提取两种方法对牡丹花粉中的黄酮类物质进行提取,以总黄酮得率为指标,通过正交试验对两种提取方法的工艺进行优化。并利用体外抗氧化测试法对两种工艺的提取粗产物进行了总抗氧化能力和还原力研究比较。结果表明,水浴回流法的最优工艺参数为:溶剂100%乙醇,提取时间60 min,水浴温度90℃,料液比1∶20(g/mL),总黄酮得率为8.28%;闪式提取法的最优工艺参数为:乙醇浓度90%,提取时间2 min,料液比1∶25(g/mL),提取转速4 000 r/min,总黄酮得率为7.89%;两种方法黄酮得率相近。另外两种方法提取产物的抗氧化性数据显示,闪式提取是一种不改变提取物活性、高效率的提取方法。
Reflux extraction water bath and Homogenate extraction are two ways to extract the Flavonoids of Peony Pollen.The extraction process was optimized by orthogonal.Antioxidant activities of Peony pollen extracts were studied.Including Trolox and reducing power.The results showed that the optimal conditions of reflux extraction water bath were: 100 % ethanol solvent,extraction time is 60 min,the temperature 90 ℃,ratio of material to liquid is 1∶20(g/mL),the extraction rate of total flavonoids was 8.28 %;the best Homogenate extraction conditions were: 90 % ethanol solvent,extraction time is 2 minutes,speed of 4 000 r/min,ratio of material to liquid is 1∶25(g/mL),the extraction rate of total flavonoid was 7.89 %;the produced rate is almost same of these two extraction methods.The result of antioxidant activities of peony pollen extractions showed that the homogenate extraction method was a way of high efficiency and without changing the activity of extraction.
