摘要
目的:研究SRC-3、IGF-1蛋白及基因在子宫肌瘤中的表达,探讨其与子宫肌瘤发生发展的关系。方法:采用免疫组织化学SP法、RT-PCR和Western blot分别从细胞、基因及蛋白水平检测SRC-3和IGF-1在子宫肌瘤及瘤旁正常组织中的表达。结果:SRC-3蛋白的阳性表达主要定位于细胞胞浆中,少数位于细胞核。IGF-1蛋白的阳性表达定位于细胞胞浆中。SRC-3、IGF-1蛋白在子宫肌瘤中的阳性表达率(67.65%,80.88%)均显著高于瘤旁正常组织(16.18%,27.94%)(P均<0.05)。SRC-3 mRNA、IGF-1 mR-NA在子宫肌瘤中的表达水平均显著高于瘤旁正常组织(P均<0.05)。子宫肌瘤中SRC-3蛋白的表达与IGF-1蛋白的表达呈正相关(r=0.303,P<0.05)。结论:SRC-3、IGF-1与子宫肌瘤的发生发展有关。SRC-3可能参与调节IGF-1介导的信号传导通路,促进子宫肌瘤的发生发展,有望为子宫肌瘤的临床治疗提供新的研究方向。
Objective:To study the expressions of SRC-3 and IGF-1 proteins in uterine leiomyoma as well as their genes,aiming at discussing the relationship between SRC-3 、IGF-1 and the development of uterine leiomyoma.Methods:Immunohistochemistry SP,RT-PCR and Western blot were used to detect the expressions of SRC-3 and IGF-1 in uterine leiomyoma and peri-tumorous normal tissues.Results:The positive expression of SRC-3 protein was mainly located in the cytoplasm,and few in the cell nucleus.The positive expressions of IGF-1 protein were found in the cytoplasm.The positive expression rates of SRC-3 and IGF-1 proteins in leiomyoma cases were 67.65% and 80.88%,which is significantly higher than those in the peri-tumorous normal tissues(16.18%,27.94%)(P0.05).The expression of SRC-3 protein was positively correlated with the expression of IGF-1 protein(r=0.303,P0.05).Conclusions:SRC-3 and IGF-1 are related with the development of uterine leiomyoma.SRC-3 may be involved in regulating the signal pathways in which IGF-1 induced,so that it accelerates the development of uterine leiomyoma,and it may be a new research direction in the clinical therapy to uterine leiomyoma.
