Expression of inflammatory factors on the vessel wall of patients with lower extremity arterial occlu- sive disease

摘要 目的定量分析下肢动脉核因子(NF)-κB和血小板源性生长因子(PDGF)的表达,探讨其在下肢动脉硬化闭塞症患者血管壁的表达及相互关系。方法对病例组19例终末期动脉硬化的下肢动脉和对照组3例正常下肢动脉NF-κB和PDGF的表达进行实时定量聚合酶链反应(RT-qPCR)分析。结果病例组患者动脉壁NF-κB(35.42±5.69)和PDGF-A(34.38±5.80)、PDGF-B(33.95±5.92)mRNA的相对表达量均高于对照组(P〈0.05),且NF-κB与PDGF-A(r=0.926)和NF-κB与PDGF-B(r=0.925)之间均存在线性正相关(P〈0.05)。结论下肢动脉硬化闭塞症患者血管壁存在NF-κB和PDGF高表达,并且两者表达相关。 Objective To analyze the expression of nuclear faetor-KB (NF-κB) and platelet derived growth factor (PDGF) on the intima and media by using quantitative analysis, and investigate the changes in the expression patterns of NF-κB and PDGF on the vessel wall of patients with lower extremity arterial occlusive disease. Methods Nineteen patients with lower extremity arterial occlusive disease served as study group, and 3 patients with other diseases as control group, respectively. The expression of NF-KB and PDGF on the intima and media of the resected arteries was detected by using real-time polymerase chain reaction (RT-qPCR). Results The mRNA expressions of NF-KB (35.42 ±5.69 ), PDGF-A (34. 38±5.80) and PDGF-B (33.95 ±5.92) on the vessel wall of study group was significantly higher than that in control group ( P 〈 0. 05 ). There was a positive linear correlation between NF-κB and PDGF-A (r=0.926) or between NF-κB and PDGF-B (r=0.925,P〈0. 05). Conclusion There was higher expression of NF-KB and PDGF on the atheroselerotic vesssel walls of patients with lower extremity arterial occlusive disease, and a positive linear correlation between NF-KB and PDGF.
