摘要 目的研究姜黄素对中波紫外线(UVB)照射HaCaT细胞凋亡的影响。方法使用MTT法测定不同浓度姜黄素对UVB照射HaCaT细胞增殖的影响;流式细胞仪测定细胞凋亡率;分光光度法检测反映出细胞内caspase-3及cas-pase-9活性的改变。结果 UVB照射HaCaT细胞后细胞增殖较未接受UVB照射组明显下降(P<0.05),细胞凋亡率可增加5%以上;对姜黄素做预处理后,UVB照射可在一定程度上抑制HaCaT细胞增殖,尤其是当浓度在10.0μmol/L时,细胞凋亡明显,caspase-3及caspase-9的活性也明显增加。结论姜黄素能增强中波紫外线诱导HaCaT细胞的凋亡,这一过程可能是通过激活caspase-3及caspase-9而实现的。 Objective To investigate the influence of curcumin on ultraviolet B(UVB) irradiation of HaCaT cells apoptosis. Methods The MTT method was used to determine the influence of different concentrations of curcumin on UVB irradiation of HaCaT cells proliferation.The flow cytometry was used to determine the cell apoptosis rate.The spectrophotometry was used to determine the changes of intracellular caspase-3 and caspase-9 activity.Results After UVB irradiation of HaCaT cells,the cell proliferation significantly reduced compared to that without UVB irradiation(P0.05) and the cell apoptosis rate could increase by over 5%.After pretreatment with curcumin,UVB irradiation could inhibit HaCaT cells proliferation to some extent,especially at the concentration of 10.0 μmol/L,where the cell apoptosis was obvious and the caspase-3 and caspase-9 activity also increased significantly.Conclusion Curcumin can enhance UVB-induced HaCaT cells apoptosis through activating caspase-3 and caspase-9 possiblely.
