摘要
目的研究乙肝病毒X蛋白(Hepatitis B virus X protein,HBx)对肝癌新标记物GP73表达的影响。方法建立稳定转染HBx基因的L02(L02/HBx)细胞系,用Real-time PCR和Western blot检测转染后GP73的表达情况。结果转染HBx基因后GP73的mRNA和蛋白表达水平明显下调,分别为对照组的(52±2)%和(42±5)%(P<0.05)。结论乙肝病毒X蛋白下调肝癌标志GP73的表达,乙肝病毒(HBV)上调GP73的表达通过其他途径实现的,是多种调控途径综合的结果。
Objective The influence and significance of Hepatitis B virus X protein to GP73 expression. Methods A Stable transfection with HBx gene cell line L02/HBX was established. The levels of mRNA and protein expression of GP73 were detected by Real-time PCR and Western Blot in control cell line L02/pcDNA3.1 and stable transfected cell line L02/HBX. Results After HBx gene transfection, the expression of GP73 mRNA level and protein level were both decreased, by 0. 52 ±0.02 and 0. 42 ± 0. 05 to the control ( P 〈 0. 05 ). Conclusion HBx decreases the expression of tumor marker GP73, HBV raised the expression of GP73 through other ways.
