HPLC-ELSD法测定酸枣仁分散片中两种成分含量的研究

摘要 目的:建立同时测定酸枣仁分散片中酸枣仁皂苷A、B成分含量的HPLC-ELSD方法。方法:应用高效液相色谱法,采用Venusil XBP-C18色谱柱(4.6mm×250mm,5μm);以乙腈(A)-水(0.1%乙酸)(B)为流动相梯度洗脱;流速1.0mL.min-1,柱温25℃,蒸发光散射检测器的漂移管温度为100℃,载气流速为2.9L.min-1。结果:酸枣仁皂苷A在1.12~11.24μg.g-1(r=0.9996)、酸枣仁皂苷B在1.36~13.56μg.g-1(r=0.9997)范围内线性关系良好,平均加样回收率(n=6)分别为:101.25%、98.63%;RSD分别为1.49%、2.57%。结论:本法快速简便,精密度好,灵敏度高,在同一色谱条件下实现多指标不同成分的同时测定,为酸枣仁分散片的全面质量评价提供参考。 Objective: To establish an HPLC-ELSD method for the simultaneous determination of jujuboside A and jujuboside B in Semen Ziziphi Spinosa dispersible tablet. Methods:The chromatographic separation was performed on a Venusil XBP reversed phase C18 (4.6mm × 250mm, 5 um ) analytical column by a gradient elution program with acetonitrile-water (containing 0. 1% acetic acid) at a flow rate of 1. 0 mL · min-1, The column temperature was 25 ℃, the evaporator tube temperature of ELSD was set at 100 ℃ with the nebulizing gas flow-rate of 2.9 L · min-1. Results:The linear range of jujubosideA and jujuboside B were 1.12-11.24 ug · g-1 (r=0. 9996) and 1.36-13.56 ug · g-1 (r=0. 9997). The average recoveries(n=6) were 101.25 % (RSD=1.49%) and 98.63% (RSD=2.57%). Conclusion: The method is simple, accurate can be used for the quality control of Semen Ziziphi Spinosae dispersible tablet.
