槲蕨愈伤组织的诱导研究 被引量:3

摘要 目的对槲蕨愈伤组织诱导条件和增殖培养条件进行了探讨。方法以槲蕨幼嫩根茎、孢子叶片和叶柄为外植体,进行了愈伤组织的诱导和增殖培养,并对愈伤组织生长量和培养过程中的褐化问题进行了初步统计分析。结果 1/2MS+2,4-D 1.0 mg/L+NAA 0.5 mg/L+6-BA 0.3 mg/L+Vc 10 mg/L为诱导槲蕨愈伤组织的较优培养基,1/2MS+2,4-D1.0 mg/L+6-BA0.5 mg/L+Vc 10 mg/L为槲蕨愈伤组织增殖培养的较优培养基。结论槲蕨幼嫩根茎能成功诱导出愈伤组织,但诱导率和增值率较低;添加Vc能较好地控制培养过程中的褐化问题,诱导培养20 d转移继代亦能较好地控制褐化的加重。 Objective To research the condition of Drynaria fortunei(Kunze) J.Sm.callus induction.Methods The young rhizome,sporophyll and petiole were used as explants to research on callus induction,and the optimum hormone constitution for callus growth and the browning in callus subculture were also studied.Results The results indicated that 1/2MS+2,4-D 1.0mg/L + NAA 0.5mg/L + 6-BA 0.3mg/L + Vc 10mg/L was the optimum medium for inducing callus,and 1/2MS + 2,4-D1.0 mg/L + 6-BA0.5 mg/L Vc 10mg/L the optimum medium for proliferation.Conclusion The callus of Drynaria fortunei(Kunze) J.Sm.young rhizome could be successful inducted,but the induction rate and multiplication rate was low.The browning in subculture could be inhibited by the medium with Vc(10 mg/L),and the subculture in the twentieth could be also inhibited.
