Effects of Physical Training from Early Age on Neuromuscular Junction Development in Postnatal Rats

摘要 目的:探讨运动对出生后早期神经肌肉连接(Neuromuscular junction,NMJ)发育的影响及其有关机制,为早期发展运动能力研究提供依据。方法:新生Sprague-Dawley雄性大鼠随机分为运动组和对照组。运动组进行每周6天、每天1小时的游泳训练,于3周龄时测定并比较运动组和对照组大鼠腓肠肌运动终板周长和终板面积、N乙酰胆碱受体(nAChR)γ-、ε-、δ-亚基、发育诱导因子Agrin及其受体MuSK以及肌源性神经营养因子NT-3表达水平。结果:运动组大鼠3周龄时腓肠肌运动终板面积和周长均显著大于对照组;腓肠肌成熟型亚基ε-AChR和诱导因子Agrin及其受体MuSK表达水平也显著高于对照组。结论:早期运动可以在一定程度上促进NMJ形态和结构成分的发育,其中发育诱导因子表达增加可能是其机制之一。 Purpose This study aimed to evaluate the effects of physical training from early age on the neuromuscular junction (NMJ) development of postnatal rats. Methods Neonatal Sprague-Dawley rats were divided into control group (C) and training group (T). Rats in group T swam 1 h per day,6 days per week from the first day to the 21th day after birth. The perimeter and area of endplate,and the expressions of γ-, ε-and δ-subunits of nacetylcholine receptor (nAChR),growth inducing factor Agrin and its receptor MUSK, and myogenic neurotrophic factor NT-3 were determined and compared 3 weeks after birth between the groups C and T. Results The perimeter and area of endplate in gastrocnemius of group T were significantly greater than that of group C. The expressions of mature ε-AChR subunit, A-grin and its receptor MuSK were higher in group T than that in group C. Conclusion Physical training from early age promoted the development of NMJ, and increased expression of growth inducing factor A-grin could be involved in this process.
