摘要
目的:中药马勃作用于体外培养大鼠皮肤的成纤维细胞,观察大鼠成纤维细胞增殖及胶原合成的能力,探讨马勃有效组分在细胞水平上促皮肤患处愈合的作用机制。方法:运用体外细胞培养技术对新生大鼠乳鼠皮肤的成纤维细胞进行培养,实验分为对照组(不给药组)和马勃有效组分37.5、75.0、150.0及300.0mg.L-1组;实验组于传代培养24h后加入不同浓度的药物,继续培养24、48、72h后收集细胞。采用MTT比色法测定细胞增殖,生物素双抗体夹心酶联免疫吸附法测定细胞胶原含量。结果:与对照组比较,37.5、75.0、150.0及300.0mg.L-1实验组的成纤维细胞增长率和成纤维细胞胶原合成能力均显著性升高(P<0.01)。在药物作用时间方面,与对照组比较,在24、48和72h观察细胞增殖率和胶原合成能力均显著增强,72h最强,差异有统计学意义(P<0.01)。结论:在不同剂量马勃有效组分作用下,大鼠成纤维细胞活化增殖,胶原合成的能力增强。
Objective To study the effects of puffball on proliferation and collagen synthesis on rat skin fibroblasts,and to investigate the mechanism of enhancing wound healing of active constituent of puffball at the cells level.Methods The rat skin fibroblasts were divided into 5 groups,all the cells were treated with puffball(37.5,75.0,150.0 and 300.0 mg·L-1),except for control group,for 24,48 and 72 h,respectively.And then MTT was used to detect the proliferation,and the content of collagen were measured by double antibody sandwich ELISA.Results Compared with control group,the cells treated with different concentrations(37.5,75.0,150.0 and 300.0 mg·L-1) of puffball showed a significant rise both in its proliferation and the ability of collagen synthesis(P0.01).Furthermore,in time-dependant manner,the proliferation rate of cells and collagen synthesis ability were significantly increased after treated for 24,48,72 h,especially in 72 h(P0.01).Conclusion Puffball with different doses can enhance the proliferation and collagen synthesis in skin fibroblasts of rats.
