鳗鲡病毒性疾病病料中鳗鲡疱疹病毒的PCR检测 被引量:20

Polymerase Chain Reaction for the Detection of Herpesvirus Anguillae in Eel Viral Disease Samples

摘要 结合流行病学、临床症状、病理变化等对2006~2012年收集的发病濒死鳗鲡病料进行分类,确定疑似鳗鲡病毒性疾病病料36份,提取病料主要内脏器官(肝、脾、肾)DNA,进行鳗鲡疱疹病毒的PCR检测。从"脱粘败血病"、"红头病"、"烂鳃病"的鳗鲡病料中检测到6份阳性结果,表明这些疾病可能是由鳗鲡疱疹病毒引起的。DNA测序结果显示,扩增的病毒DNA聚合酶基因片段与分离自台湾、日本及欧洲的病毒株完全一致,表明这些病毒可能为同一病毒株。 Diseased eels collected between 2006- 2012 were classified by pidemiological, clinical symptoms and pathological characteristics, and 36 samples suspected to be infected by virus were identified. Total DNA was extraced from liver, spleen or kidney, and used for herpesvirus anguillae (HVA) detection. The results showed that herpesvirus anguillae were detected from 6 samples, and the disease of "Skin Mucous Debonding', "Septicemia", "Red Head", "Gill Rotten", which causing huge losses to the eel aquaculture industry, might caused by HVA; the amplified DNA sequence were exactely the same with the sequence from the starin of Taiwan, Japan and Europe, which means they might be the identical strain.
