摘要 目的探讨SOX-2及OCT-4表达与宫颈鳞癌病理分级的关系及其在宫颈癌发生、发展等方面的作用和意义。方法应用免疫组织化学技术和RT-PCR方法在蛋白及核酸水平检测SOX-2及OCT-4在正常宫颈及宫颈鳞癌组织中的表达。结果 (1)免疫组化染色显示,正常宫颈组织与中、低分化宫颈鳞癌组织中SOX-2的表达差异有显著性(P<0.05);OCT-4在正常宫颈和宫颈鳞癌组织表达差异有显著性(P<0.01)。(2)RT-PCR示SOX-2mRNA在正常宫颈及中、低分化宫颈鳞癌组织中的表达差异有显著性(P<0.01),并且SOX-2的表达随着病理学分级的下降而表达上调;OCT-4mRNA在正常宫颈及宫颈鳞癌组织中的表达差异有显著性(P<0.01),但OCT-4的表达与病理学分级无关。结论 SOX-2和OCT-4可能在宫颈鳞癌的发生、发展中起重要作用。 Objective To investigate the effect and significance of SOX-2 and OCT-4 in tumorigenesis,development and the relationship between the expression of normal cervix and the pathological grade in cervical carcinoma.Methods The expression of protein and nucleic acid levels of SOX-2 and OCT-4 in normal cervix and cervical squamous cell carcinoma were detected by immunohistochemistry and RT-PCR.Results(1) IHC showed the difference was significant(P0.05) in the expression of SOX-2 protein in middle and poorly differentiated cervical squamous cell carcinoma compared with those in normal cervix and well-differentiated cervical carcinoma.The significant differences(P0.01) were found in normal cervix and cervical squamous carcinoma,but the expression of OCT-4 do not following up of pathological grades.(2) RT-PCR showed that the expression of the SOX-2 mRNA was found significant differences in normal cervix and poorly differentiated cervical squamous carcinoma.The result demonstrate that the expression of SOX-2 following the decline of pathological grades.The significant differences(P0.01) were found in normal cervix and cervical squamous carcinoma.The expression of OCT-4 mRNA was found significance difference between normal cervix with cervical carcinoma.The result indicates that expression of OCT-4 was significantly enhanced at cervical carcinoma and the difference between each pathological grade was not significant.(P0.01).Conclusion SOX-2 and OCT-4 may play an important role in the development of cervical squamous cell carcinoma.
