摘要
目的 观察 15日龄大鼠小脑 ,海马 ,嗅脑一氧化氮 (NO)含量 ,一氧化氮合酶 (NOS)活性的变化及甲状腺激素对上述部位神经元型一氧化氮合酶 (nNOS)基因表达的调节。方法 采用丙基硫氧嘧啶 (PTU)给孕母鼠灌胃造成仔鼠甲减动物模型 ;采用NO ,NOS生化测定法及nNOSmRNA半定量逆转录聚合酶链反应 (RT PCR)法。结果 无论正常组还是甲减组 ,NO含量 ,NOS活性及nNOSmRNA转录均以小脑为最高 ,嗅脑次之 ,海马最低 (P <0 .0 5 ) ;正常组NO含量 ,NOS活性nNOSmRNA转录均高于甲减组 (P <0 .0 1)。结论 提示甲状腺激素对nNOS基因表达有上调作用 ,NO信号系统可能参与大鼠小脑 ,海马 ,嗅脑等区甲状腺激素缺乏所造成的脑损害过程。
Objective To observe nitric oxide (NO) contents, nitric oxide synthase (NOS) activities and nNOS mRNA gene expressions in cerebellum, olfactory bulb and hippocampus of rats at the age of 15 days. Methods Perinatal hypothyroidism was induced by infusing PTU solution to the dams (1%PTU 0.02 ml/g) by gavagy for 5 8 days beginning on embryonic days 15. Biochemistry measurement of NO and NOS and semi quantitative reverse transcription polymerase chain reaction (RT PCR) analysis of nNOS mRNA were used. Results In both hypothyroidism rats and normal controls, the contents of NO, activities of NOS and nNOS mRNA gene transcription levels (corrected by GAPDH mRNA) were highest in cerebella, higher in olfactory bulbs, and lowest in hippocampuses. The contents of NO and the activities of NOS were lower in hypothyroidism rats than those in controls and nNOS mRNA dropped also significantly in hypothyroidism rats as compared to that in normal controls. Conclusion The thyroid hormone up regulates the expression of nNOS gene and NO signal conduct system may take part in the course of impairment in the brain development induced by thyroid hormone deficiency.
