摘要
目的探讨更年宁神汤对围绝经期大鼠卵巢颗粒细胞caspase-3蛋白的影响。方法选用雌性wister大鼠40只,月龄13~15月,随机分为4组：模型组、更年宁神汤高剂量组、更年宁神汤低剂量组、西药利维爱组,每组各10只。另选5月龄、体质量（180±20）g的雌性wister大鼠10只作为青年对照组。中药高剂量组灌服更年宁神汤5.68 g/（kg.d）,中药低剂量组更年宁神汤1.42 g/（kg.d）,围绝经期模型组灌服等量生理盐水,西药对照组每日灌服3mL西药利维爱溶液0.26 mg/（kg.d）,连续治疗4周。采用免疫组织化学法及Westernblot方法检验各组大鼠卵巢颗粒细胞caspase-3蛋白的表达。结果模型组大鼠卵巢中caspase-3蛋白表达高于青年对照组,治疗组caspase-3蛋白表达水平较模型组降低,更年宁神汤高剂量组、利维爱组大鼠卵巢中caspase-3蛋白表达低于模型组差异显著;中药高剂量组与中药低剂量组比较有统计学意义;中药高剂量组与利维爱组比较比较,差异无统计学意义。结论更年宁神汤治疗围绝经期综合征的机制可能是通过抑制卵巢颗粒细胞分泌caspase-3凋亡因子,延缓卵巢衰老,疗效与利维爱相当。
Objective： To study the effects of Gengnianingshen Decoction（GNNSD） on expression of caspase-3 of ovarian granulose cell in rats during perimenopausal period.Methods： Forty 13~15 week-old female SD rats were randomly divided into 4 groups（the GNNSD low dose and high dose groups,the livial group and aged control group）.Ten young female rats were selected as young control group.The high dose groups were given Gengnianingshen Decoction 5.68 g/（kg·d）,low dose groups were given Gengnianingshen Decoction 1.42 g/（kg·d）.The model group group were given normal saline with the same dose.The control group were given livial solution.The expression of capase-3 protein of ovarian granulose cell were detected by immunohistochemistry and Western blot.Results： The levels of caspase-3 protein in rat ovaries was up regulated in the control group compared to normal group,the expression of caspase-3 protein in GNNST high dose group was down regulated compared to the control group and GNNST low dose group;the levels of caspase-3 protein showed no difference between GNNST high dose and livial group.Conclusion： GNNST high dose formulae can down-regulate the expression of caspase-3 protein in rat ovaries of during perimenopausal period.It may be one of the molecular mechanisms of GNNST postponed the ovarian failure and cured perimenopausal syndrome.
