摘要
目的探讨山茱萸总苷对急性缺氧乳鼠心肌细胞凋亡的影响。方法原代培养乳鼠的心肌细胞,采用液状石蜡封闭法建立缺血缺氧模型,并将原代培养3d的心肌细胞分为对照组、缺氧组(1h、2h、3h、5h)及治疗组,药物浓度为生药10g/L,采用TUNEL法检测各组的凋亡率。结果与对照组相比,缺氧组各时间点凋亡率明显上升,差别有统计学意义(P<0.01);与缺氧组比较,治疗组各个时间点的凋亡率都有所下降(P<0.01)。结论缺氧可以引起心肌细胞凋亡,并且随着缺氧时间的延长凋亡率上升,山茱萸总苷可以抑制心肌细胞的凋亡。
Objective To investigate the effects of cornus officinalis glycosides(COG) on myocardiocyte apoptosis in suckling mouse with acute hypoxia.Methods Myocardiocytes of suckling mouse were incubated with liquid paraffin closed method to establish the ischemic anoxia model.The generation of 3 days training myocardial cells were divided to normal group,hypoxia group(1 h,2 h,3 h,5 h),and COG group.The concentration of COG was 10g/L as the crude drug,and apoptosis rate was studied by TUNEL method.Results Compared with normal group,the apoptosis rate in hypoxia groups were increased obviously(P0.01).The apoptosis rate in COG groups were decreased compared with that in hypoxia group(P0.01).Conclusion Acute hypoxia could cause the apoptosis of myocardial cells,and the apoptosis rate was increased with the extension of anaerobic time.COG could inhibit the apoptosis of myocardial cells in suckling mouse with acute hypoxia.
