摘要
目的:建立密脉鹅掌柴提取物中总皂苷及齐墩果酸的含量测定方法。方法:总皂苷的含量测定采用比色法,于552nm测定;齐墩果酸的含量测定采用高效液相色谱法,色谱柱:Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm),流动相:乙腈-水(84:16),流速:1.0 ml·min^(-1),检测波长:203 nm,柱温:25℃。结果:按选定的紫外条件进行测定,齐墩果酸在2.0~20.0μg·ml^(-1)范围内呈良好线性关系(r=0.999 5),平均回收率为99.57%;HPLC定量试验中,齐墩果酸在0.4~8.0μg范围内呈良好线性关系(r=0.999 9),平均回收率为100.44%。结论:建立的方法简便易行,结果准确可靠,可作为本产品的质量控制方法。
Objective:To establish the method for determination of total saponins and oleanolic acid in Schefflera vemdosa extracts. Method:The content of total saponins was detennined by UV at 552 nm.The separation and determination of oleanolic acid were performed on an Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm) column with the mobile phase consisted of acetonitrile-water (84:16) at the flow rate of 1.0 ml ? min^(-1),the detection wavelength was 203 nm,and the column temperature was 25X1.Result: The standard curve was linear within the range of 2.0-20.0μg ? ml^(-1) for oleanolic acid of UV,0.4-8.0 u,g for oleanolic acid of HPLC.The average recovery of total saponins and oleanolic:acid was 99.57%and 100.44%,respectively.Conclusion:The developed methods are rapid and accurate with high repeatability and stability,and can be used in the quality control of Schefflera venulosa extracts.
