摘要
目的 :建立简单、有效的猪血小板衍生生长因子 (p PDGF)纯化方法。方法 :采用超声裂解、酸醇抽提、离子交换色谱、凝胶色谱等方法提纯 p PDGF,用 SDS-聚丙烯酰胺凝胶电泳测定其相对分子质量 ,并检测其生物学活性。结果 :纯化的 p PDGF相对分子质量为 (2 .879～ 3.0 78)× 10 4,纯化倍数为 75 82倍 ,活性回收率为 3% ,比活性为 3.41176× 10 5 U/ mg。 结论 :提取和纯化 p PDGF的方法简单 ,且达到较好的结果 ,为进一步研究 PDGF的理化性质及生物学特性打下基础。
Objective: To prepare the highly purified porcine platelet derived growth factor (pPDGF) from porcine platelets with simple and effective method. Methods: pPDGF had been purified from porcine platelets by acid ethanol extraction, ion exchange chromatography and gel chromatography. The molecular mass was measured by SDS PAGE. Results: The purified pPDGF had a molecular mass ranging from (2.879～3.078)×10 4, the fold of purification of pPDGF was 7 852 and recovery and specific activity was 3% and 3.411 76×10 5 U/mg respectively. Conclusion: A simple and effective purification method for pPDGF is established. It is the basis for further researching the biology of PDGF.
