摘要
目的 研究 SDS-聚丙烯酰胺凝胶电泳 (SDS- PAGE)显示小分子多肽的方法 .方法 观察不同方法处理的样品 ,上样量对电泳结果的影响及对分子量标准 (Mr 2 5 12~ 16 949)进行直线回归分析 .结果 样品的不同处理条件未见有差异 ;在该实验系统条件下上样样品为每孔 5~ 10μg较佳 ;分子量标准直线回归系数 r=- 0 .96 2 .结论 样品处理方便 ;上样量少 ;在 16 0 g· L- 1 T,6 0 g· kg- 1 C较低的丙烯酰胺含 6 mol· L- 1脲的分离胶中能够显示 Mr为 2 5 12的多肽 。
AIM To establish a sensitive method of electrophoresis for low molecular weight peptides. METHODS Tricine SDS polyarcrylamide gel electrophoresis with 6 mol·L -1 urea was used. The samples were treated with different methods(temperature and time interval) and varying loading quantity of samples. The linear regression analysis was made. RESULTS Different treatment of samples had no effect on the experimental results. The optimum loading scope of samples was 5~10 μg per lane. The correlation coefficient of molecular markers was -0.962. CONCLUSION Separating gel with 6 mol·L -1 urea at lower acrylamide concentration(160 g·L -1 T, 60 g·kg -1 C) is useful for identifi cation of peptide with Mr from 2512 to 16949. It is convenient and simple to operate and is a well established method for analysis of low molecular peptides.
