茉莉酸甲酯调控阳春砂HMGR、DXR和DXS基因表达 被引量:12

Regulatory Effect of Methyl Jasmonate on HMGR,DXR and DXS Genes Expression in Amomum villosum Lour.

摘要 【目的】探讨茉莉酸甲酯(Methyl Jasmonate,MeJA)调控阳春砂萜类化合物生物合成途径3个关键酶基因HMGR、DXR、DXS表达的规律。【方法】用20μmol/L和200μmol/L高、低2个浓度的MeJA喷施阳春砂幼苗,诱导不同时间后取样,提取RNA,采用荧光定量PCR法测定AvHMGR、AvDXR、AvDXS相对表达量。【结果】MeJA对AvHMGR、AvDXR和AvDXS的表达均有促进作用,其中对AvDXR的促进作用最明显,AvHMGR次之,两者最高表达量分别达到处理前的9.40和3.95倍;AvDXR基因受高、低浓度MeJA的诱导表达均十分明显,而AvHMGR基因仅对低浓度MeJA敏感,AvDXS基因对高、低浓度的MeJA均不敏感;AvHMGR和AvDXR的表达量在诱导后24 h达到最高峰,在72 h恢复到处理前水平。【结论】诱导子MeJA对AvDXR和AvHMGR基因表达的调控是激发式的,基因表达迅速上调到一定水平,而后逐渐回落。 Objective To investigate the rule of methyl jasmonate(MeJA) in regulating the expression of HMGR,DXR and DXS,which are the genes encoding the key enzymes of terpenoids biosynthetic pathway in Amomum villosum Lour..Methods The seedlings of A.villosum were given spraying with 20 and 200 μmol/L MeJA.After inducing with MeJA for different time,RNA was extracted from the sample leaves.Expression levels of AvHMGR,AvDXR and AvDXS genes were detected by real-time quantitative PCR.Results The expression of AvHMGR,AvDXR and AvDXS was up-regulated by the induction of MeJA.The expression level of AvDXR was the highest,accounting for 9.40 folds of that before treatment,and AvHMGR came next,accounting for 3.95 folds of that before treatment.AvDXR was induced significantly by both high-and low-concentration MeJA.However,AvHMGR was only sensitive to the induction of low-concentration MeJA.AvDXS was insensitive to the induction of either high-or low-concentration MeJA.The expression of AvHMGR and AvDXR reached the highest level at hour 24,and then recovered to the former level at hour 72 after the induction.Conclusion MeJA shows an excitation style in regulating AvDXR and AvHMGR expression,which is characterized as first rapid up-regulation and then slowlg coming back to the former level.
