鸭坦布苏病毒JS2010株全基因组的克隆及序列分析 被引量:2

Cloning and sequence analysis of the complete genome of duck Tembusu virus JS2010 strain

摘要 为进一步认识鸭坦布苏病毒(DTMUV)的分子特征,了解该病毒的遗传进化关系,根据GenBank上登录的DTMUV基因组序列设计并合成了11对引物,通过RT-PCR方法对DTMUV JS2010株的基因组序列进行分段扩增并测序。结果显示,该病毒全基因序列长10 990nt(GenBank登录号:JX273153),具有一个10 278nt的蛋白编码序列。全基因组序列分析表明,JS2010株与DTMUV参考毒株的核苷酸序列同源性为99.2%~99.6%,氨基酸序列同源性为99.2%~99.8%。E基因序列比对和系统进化树分析发现JS2010株与黄病毒属Ntaya病毒群的Tembusu病毒的同源性为87.7%~99.5%,与同属的Bagaza病毒、Ntaya病毒的同源性分别为70.5%和75.6%,表明JS2010株与Ntaya病毒群,特别是与Tembusu病毒有很高的亲缘关系。 The complete genomic sequence of duck Tembusu virus(DTMUV) JS2010 strain was amplified by RT-PCR using 11 pairs of primers designed according to the published sequence of DTMUV deposited in GenBank in order to investigate its molecular structure and genetic characteristics.Sequence analysis showed that the complete genome of DTMUV JS2010 was composed of 10 990 nucleotides in length(GenBank accession No.JX273153),which included a 10 278 nt single large open reading frame encoding a polyprotein.The JS2010 strain shared from 99.2% to 99.6% nucleotide homology and shared from 99.2% to 99.8% amino acid identity with other DTMUV strains.The sequenced fragment of E gene of the JS2010 strain shared from 87.7% to 99.5% nucleotide identity with the Tembusu virus of Ntaya virus group,and the nucleotide identity was seen with the Bagaza virus strain(70.5%) and with the Ntaya virus strains(75.6%).All results showed that the JS2010 strain shared the highest similarity with Tembusu virus of Ntaya virus group.
