牛磺酸联合血管紧张素转化酶抑制剂对大鼠急性肺损伤的保护作用 被引量:1

摘要 目的探讨牛磺酸及血管紧张素转化酶抑制剂对大鼠急性肺损伤(ALI)的保护作用及其相关机制。方法将130只大鼠随机分成5组,正常对照组(n=10)、肺损伤组,牛磺酸组,卡托普利组,牛磺酸和卡托普利组(n=30)。静脉注射脂多糖5 mg/kg,建立ALI模型,脂多糖预激12 h后分别注射牛磺酸50 mg/kg,卡托普利1.25 mg/kg及两者联用,于注射后3、6、9、12、24 h时间点采集标本,分别进行肺组织病理学观察,并采用ELISA法分别检测血浆及肺泡灌洗液中的可溶性细胞间黏附分子-1(sICAM-1)、内皮素-1(ET-1)、血管性血友病因子(vWF)的水平。结果肺损伤组病理组织切片显示炎症和肺水肿现象,肺泡间隔明显增厚,肺泡腔变窄。牛磺酸组、卡托普利组及两药联用组能使上述病理变化改善sICAM-1、ET-1、vWF在各肺损伤组中较对照组均有所升高,在牛磺酸治疗组、卡托普利治疗干预后均有所降低,并以两者联合干预效果最明显。结论牛磺酸和血管紧张素转化酶抑制剂的联合干预能使肺组织病理变化较肺损伤组及单药干预组明显减轻,且sICAM-1、ET-1、vWF明显降低,因此提示二药联合对肺血管内皮细胞具有保护作用。 Objective To investigate the protective effect by taurine and angiotensin converting enzyme inhibitor on acute lung injury (ALl) and its relative mechanism. Methods 130 rats were randomly divided into five groups:normal control (n = 10) ,lung injury, taurine,captopril,taurine and captopril groups (n = 30). ALI model was established by injection of LPS 5 mg/kg. After pre-activating for 12 hours,the taurine 50 mg/kg, captopril 1.25 mg/kg, and both combined were injected and then, at 3,6,9,12,24 h time point 'after injection, specimens were collected ,respectively. Lung histopathology was observed and analyzed. Soluble intercellular adhesion molecule -1 (sICAM- 1 ) , endothelin-1 ( ET-1 ) ,von Willebrand factor (vWF) levels were detected in plasma and bronchoalveolar lavage fluid by ELISA. Results Lung injury group showed marked inflammation and mild lung edema with marked thickening of alveolar septa and alveolar space narro- wing. Improved the pathological changes were observed in the taurine, captopril and both combined groups, sICAM-1, ET-1, vWF were increased in lung injury group than those of control group. But after the intervention, they were lower in taurine, captopril groups, and two joint intervention was most obvious. Conclusions Intervention of taurine and angiotensin-converting enzyme iuhibitors combined could make inju- ry in lung tissue biopsy significantly reduced compared with the Lung injury group and a single drug-intervention group, sICAM-1 ,ET-1, vWF are significantly lower and therefore it prompts that two-drug combination has a protective effect on pulmonary vascular endothelial ceils.
