摘要
目的:观察P53基因在甲减大鼠肾脏中的表达变化,探讨凋亡在甲减肾损伤发病过程中的作用。方法:本研究在复制甲减大鼠模型的基础上,利用RT-PCR方法分析肾脏组织中P53mRNA的表达变化。结果:甲减组(HYPO)与对照组(CL)比较,血清TSH明显上升(P<0.05),TT3、TT4、FT3、FT4均较前明显下降(P<0.01),表示甲减模型复制成功。甲减大鼠肾脏组织中P53mRNA表达明显下降(P<0.05)。结论:低碘摄入可导致甲减,甲减会引起P53基因表达下降,进而导致肾脏细胞凋亡增加,引起肾脏损伤。
Objective:To observe the expression of P53 in hypothyroid rat kidney,study the effect of P53 on hypothyroid renal damage Methods:At the base of animal models of hypothyroidism,to observe the gene expression of P53 by using RT-PCR.Results:Compared with CL rat,TSH was significantly increased(P0.05),TT3,TT4,FT4 and FT3 were significantly decreased(P0.01),the levels of P53 were significantly decreased(P0.05) in the HYPO rats.Conclusion:Hypothyroid rat models can be produced successfully by being fed with low-iodine diet and drunk deionized water.Hypothyroidism can lead to lower gene expression of P53 in kidney.This finding suggests that apoptosis might induce hypothyroid renal damage.
