摘要
制备包裹白藜芦醇的柔性纳米脂质体,并优化配方,得到一个稳定性好、负载量高的体系。实验采用注入法制备柔性纳米脂质体;制备出的柔性纳米脂质体采用光子相关光谱(PCS)和激光衍射(LD)测得粒径为250nm~300nm,PDI为0.1~0.2;透射电镜观察出的脂质体形貌呈球形或椭球形;用高效液相色谱测出白藜芦醇的含量为0.7%~1.1%,采用超滤离心法测得的包封率为99.7%。体系于4℃、25℃以及40℃下分别保存2个月,粒径无明显变化。采用注入法制备柔性纳米脂质体,操作简便,制备出的脂质体性状稳定。
In this paper, a flexible nanoliposome containing resveratrol was prepared and a stable formulation with high content was achieved on optimized condition. In the experiment, injection method was used to prepare the nanoliposome; the particle size and polydispersity index value (PDI) of these flexible nanoliposome, tested by photon correlation spectroscopy (PCS) and laser diffractometry(LD), were 250 nm-300 nm and 0.1-0.2 respectively. The appearance of these nanoliposome were found as round or elliptical under transmission electron microscope. The concentration of resveratrol ranged from 0.7% to 1.1%, detected by high performance liquid chromatography (HPLC); and the resveratrol entrapment capacity was 99.7%. The samples have been conserved at 4℃, 25℃ and 40℃ respectively for 2 months, and the particle sizes changed little. These flexible nanoliposome containing resveratrol possessed high stability and were simple to prepare using injection method.
