摘要
目的观察倒卵叶五加及其复方对亚急性衰老大鼠血清丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性、总抗氧化能力(T-AOC)以及睾丸组织中NO、iNOS含量的影响。方法 60只雄性SD大鼠随机分为正常对照组、模型组、倒卵叶五加组、佳蓉片组和维生素E组,每组12只。D-半乳糖连续腹腔注射建立亚急性衰老大鼠模型,各组大鼠在腹腔注射D-半乳糖(正常组动物注射等量生理盐水)的同时,正常和模型组大鼠灌胃生理盐水[10 mL/(kg.d)]、其他各组依次给予倒卵叶五加[1 g/(kg.d)]、佳蓉片[6 g/(kg.d)]、维生素E[20 mg/(kg.d)],用药50 d。分别检测各组大鼠血清MDA含量、GSH-Px活性、SOD活性及T-AOC。结果①与正常对照组大鼠比较,模型组大鼠MDA含量明显升高,GSH-Px活性、SOD活性、T-AOC明显降低(P<0.01或P<0.05);睾丸组织NO和iNOS含量明显升高(P<0.01)。②与模型组比较,倒卵叶五加、佳蓉片、维生素E三组大鼠血清MDA、睾丸组织NO和iNOS含量明显降低(P<0.01或P<0.05),GSH-P活性、SOD活性、T-AOC明显升高(P<0.01)。结论倒卵叶五加和佳蓉片可升高衰老大鼠的抗氧化能力,具有明显延缓衰老的作用;机制可能与其抗氧化和降低衰老大鼠睾丸组织中NO和iNOS的含量有关。
Objective To observe the effect of Acanthopanax obovatus' Hoo and its compound preparation on malondi- aldehyde (MDA) content, glutathione peroxidase (GSH-Px) activity, superoxide dismutase (SOD) activity, the total an- tioxidant capacity in serum and NO content, iNOS content in testis of sub-acutely aging rats. Methods 60 male SD rat were randomly divided into 5 groups: normal control group, model group, Aconthopanax obouatus Hoo (AOH) group, Jiarong Tablet group and Vitamin E group, 12 rats in each group. The subacute senile rats model was made by in- traperitoneal injection (ip) of D-gal continuous. Each group was deah with D-gal except the normal group which were injected with isodose physiological saline, meanwhile the normal control group and the model group were lavaged with saline [10 mL/(kg, d)], the other group were lavaged with AOH[1 g/(kg. d)], Jiarong Tablet [6 g/(kg. d)] and Vitamin E [20 mg/(kg .d)] respectively for 50 days. The MDA content, GSH-Px activity, SOD activity, and T-AOC in serum of rats in each group were respectively detected. Results Compared with rats in normal control group, MDA content in- creased obviously, GSH-Px activity, SOD activity and T-AOC of rats in model group decreased obviously (P 〈 0.01or P 〈 0.05)the NO content and the iNOS content in testis increased obviously (P 〈 0.01). Compared with rats in model group, the MDA content in serum, the NO content and i NOS content in testis decreased significantly, GSH-Px activ- ity, SOD activity and T-AOC of rats in AHO group, Jiarong Tablet group, Vitamin E increased significantly (P 〈 0.01). Conclusion AOH and Jiarong Tablet can inhibit declining of anti-oxidative ability of subacute senile rats, has obvi- ously anti-aging effects, the mechanism maybe related with its ability of antioxidant and the effects of reducing NO, i- NOS content in testis of rats.
