摘要
目的探讨姜黄素对胰岛素抵抗小鼠脂联素表达的影响。方法将64只雄性7周龄C57BL/6小鼠随机分为正常对照组、模型对照组、50 mg.kg-1.d-1姜黄素组和250 mg.kg-1.d-1姜黄素组,每组16只。正常对照组和模型对照组小鼠每天以1.0%羧甲基纤维素溶液灌胃;姜黄素组小鼠每天以不同剂量(50 mg.kg-1.d-1和250 mg.kg-1.d-1)姜黄素灌胃;干预11周;第11周,模型对照组和姜黄素组每天同时腹腔注射肿瘤坏死因子α(TNF-α)(7μg.kg-1.d-1),连续处理8 d。检测小鼠空腹血糖和血清胰岛素,计算胰岛素作用指数(IAI);ELISA法检测血清脂联素水平;实时定量PCR测定腹部皮下脂肪组织和睾周脂肪组织中脂联素mRNA的表达。结果模型对照组小鼠IAI显著低于正常对照组(P<0.05),250 mg.kg-1.d-1姜黄素组小鼠IAI显著高于模型对照组(P<0.05)。与正常对照组比较,模型对照组小鼠血清脂联素的质量浓度显著降低(P<0.05);与模型对照组比较,250 mg.kg-1.d-1姜黄素组小鼠血清脂联素的质量浓度明显升高(P<0.05)。与模型对照组比较,50 mg.kg-1.d-1姜黄素组和250 mg.kg-1.d-1姜黄素组小鼠腹部皮下和睾周脂肪组织脂联素mRNA的表达均显著升高(P<0.05)。结论姜黄素对胰岛素抵抗小鼠有一定的保护作用,其机制可能是上调脂联素mRNA表达。
Objective To investigate the effect of curcumin on expression of adiponectin in mice with insulin resistance. Methods Sixty-four 7-week-old male C57BL/6 mice were randomly divided into normal control group, model control group, 50 mg?kg-1?d-1 curcumin group and 250 mg·kg^-1·d^-1urcumin group, with 16 mice in each group. Mice in normal control group and model control group were daily gavaged with 1.0% carboxymethyl cellulose solution for 11 weeks, and mice in 50 mg·kg^-1·d^-1urcumin group and 250 mg·kg^-1·d^-1roup were gavaged with 50 mg·kg^-1·d^-1urcumin and 250 mg·kg^-1·d^-1urcumin respectively for 11 weeks. On the eleventh week, mice in model control group, 50 mg·kg^-1·d^-1urcumin group and 250 mg·kg^-1·d^-1urcumin group were simultaneously intraperitoneally injected with tumor necrosis factor-α (TNF-α)(7 μg·kg-1·d-1)for consecutive 8 d. The fasting blood glucose and serum insulin were measured, the insulin action index (IAI) was calculated, the serum adiponectin was determined by ELISA, and the expression of adiponectin mRNA in abdominal subcutaneous adipose tissues and periepididymal adipose tissues was detected by quantitative Real-Time PCR. Results IAI in model control group was significantly lower than that in normal control group (P〈0.05), and IAI in 250 mg·kg^-1·d^-1urcumin group was significantly higher than that in model control group (P〈0.05). The mass concentration of serum adiponectin in model control group was significantly lower than that in normal control group (P〈0.05), and the mass concentration of serum adiponectin in 250 mg·kg^-1·d^-1urcumin group was significantly higher than that in model control group (P〈0.05). The expression of adiponectin mRNA in abdominal subcutaneous adipose tissues and periepididymal adipose tissues in 50 mg·kg^-1·d^-1urcumin group and 250 mg·kg^-1·d^-1urcumin group was significantly higher than that in model control group (P〈0.05). Conclusion Curcumin has definite protective effect on mice with insulin resistance, and the mechanism may be the upregulation of expression of adiponectin mRNA.
