乙醇对大肠杆菌碱性磷酸酶活性与构象的影响 被引量:1

摘要 常用有机试剂乙醇作为效应物对大肠杆菌碱性磷酸酶(EAP)活性影响状态作用机制进行了研究.结果表明,EAP酶活性随着乙醇浓度增大而迅速下降,说明乙醇对EAP有明显的失活作用,IC50为13%.关于乙醇对EAP失活作用机制的研究表明,乙醇对EAP的失活作用是以非竞争性机制模式进行的.应用荧光光谱研究EAP经乙醇微扰后的分子构象变化,发现EAP的内源荧光强度随乙醇浓度增大而增强,说明酶蛋白分子中的荧光发色基团所处微环境已经发生变化,乙醇对EAP分子构象存在显著影响. The enzymatic activity of Escherichia coli alkaline phosphatase(EAP) decreased sharply with increasing ethanol concentration. Ethanol was a non-competitive inhibitor. Fluorescence spectrum assay showed increased fluorescence intensity with increasing ethanol concentration, suggesting changed micro- conformation of Trp/Tyr residues in EAP.
