摘要
目的探讨柚皮苷对糖尿病心肌病大鼠心肌组织核因子KB(NF—KB)炎症信号通路的影响,初步阐明柚皮苷对糖尿病心肌病的防护作用机制。方法高脂高糖喂养和-次性腹腔注射链脲佐菌素(STZ)建立糖尿病心肌病大鼠模型,完全随机分为模型对照组(糖尿病心肌病组,9只)、柚皮苷治疗组(9只)和正常对照组(10只)。糖尿病大鼠模型成功后,柚皮苷治疗组予Naringin粉末40mg/kg灌胃,正常对照组和糖尿病心肌病组则按体重予蒸馏水灌胃。通过免疫组织化学法测定心肌组织NF—KB蛋白的表达,放射免疫分析法分别检测外周血肿瘤坏死因子仪(TNF-a)、白细胞介素6(IL-6)、IL-1p含量,透射电镜观察心肌组织超微结构。结果与正常对照组比较,糖尿病心肌病组大鼠心肌组织NF—KB蛋白的阳性平均光密度以及外周血TNF-a、IL-6、IL-1p分泌明显增加(均P〈0.05),柚皮苷治疗后,NF—KB蛋白、TNF-a、IL-6和IL.1B水平明显下降[(0.162±0.013)μg/L比(0.222±0.018)μg/L,(1.35±1.02)μg/L比(1.98±1.37)μg/L,(15.8±14.6)μg/L比(35.8±21.1)μg/L,(0.14±0.15)μg/L比(0.27±0.23)μg/L,均P〈0.05]。与正常对照组比较,糖尿病心肌病组大鼠心肌超微结构损伤明显;柚皮苷治疗组心肌超微结构损伤减轻。结论柚皮苷可减轻糖尿病心肌病心肌的损伤,其机制可能与其抑制糖尿病心肌病大鼠心肌组织的NF—KB信号炎症通路有关。
Objective To investigate the effects of naringin on nuclear factor ( NF)KB inflammatory signa ling pathway in diabetic cardiomyopathy rats, and to elucidate its cardioprotect mechanism in diabetic cardiomyopa thy. Methods Diabetes cardiomyopathy rat model was established by high fat and sugar feed and onetime intraper itoneal injection of low dose streptozotocin. They were divided into model control(MC) group and naringin treatment group; the other set of normal control(NC) group. Expression of NFKB in myocardium by immunohistochemistry and tumor recrosis factor a (TNF-a), interleukin (IL)6, IL-1β in serum by radio immunoassay were detected. Results Compared with the control group, positive vascular density of NFKB in myocardium and TNF-a, IL-a, IL-1β in serum were significantly increased in diabetic cardiomyopathy group, and the level of NFKB in myocardium and the level of TNF-a, IL-a, IL-1β in serum was decreased in naringin treatment group [ (0.162 :l: 0.013 )μg/L vs (0.222 ± 0. 018 ) μg/L, ( 1.35 ± 1.02) μg/L vs ( 1.98 ± 1. 37 ) μg/L, ( 15.8 ± 14. 6 ) μg/L vs ( 35.8 ± 21.1 ) μg/L, (0.14 ±0.15) μg/L vs (0.27 ±0.23)μg/L, all P 〈0.05]. Electron microscopy results showed that myocardial uhrastructure was significantly injured. Naringin pretreatment alleviated myocardial ultrastructure changes with regular myofibrillar organization, intact membrane and cristae. Conclusion Naringin can ameliorate myocardi al ultrastructure in diabetic cardiomyopathy;the mechanism is possible related to NFKB inflammatory signaling path way in diabetic cardiomyopathy rats.
