摘要 目的探讨肝泰胶囊对大鼠乙醇性肝纤维化的治疗作用及其可能的机制。方法将42只SD雄性大鼠随机分为正常对照组、乙醇模型组、肝泰治疗组。采用光镜、电镜观察各组肝组织病理学改变,Masson染色观察各组肝纤维化程度。采用放射免疫法测定血清透明质酸(HA)、Ⅲ型前胶原(PC-Ⅲ)、Ⅳ型胶原(Ⅳ-C)的含量,同时用半自动生化仪检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、T-CHOL(总胆固醇)、TG(甘油三酯);分光光度仪检测丙二醛(MDA),超氧化物歧化酶(SOD),还原型谷胱甘肽(GSH),γ-谷氨酰转肽酶(γ-GT),以及免疫组化检测α-平滑肌肌动蛋白(α-SMA)的表达。结果治疗组与模型组相比:①肝组织肝纤维化程度明显改善;②肝组织胶原纤维面积明显减少;③血清透明质酸、谷丙转氨酶、总胆固醇、γ-谷氨酰转肽酶水平显著降低(均P<0.05);④肝功能损伤明显减轻。结论肝泰胶囊对乙醇性肝纤维化有一定的治疗作用,能显著改善大鼠乙醇性肝组织损伤,减轻肝纤维化。 Objective To evaluate the efficacy of Gantai capsules in the treatment of alcohol-induced liver fibrosis in rats and explore the underlying mechanism. Methods A total of 42 rats were randomly divided into control,model and Gantaitreated groups. The hepatic tissues were histopathologically observed under the light and electron microscope. Hepatic fibrosis was evaluated by Masson Trichrome staining and the expression levels of PC-III, HA, IV-C were detected in the serum by using radioimmunoassay. The levels of AST, ALT,T-CHOL,TG,γ-GT, MDA,SOD and GSH were determined by using spectrophotometry. The expression of α-SMA in liver tissue was measured by using immunohistochemistry. Results Hepatic fibrosis was significantly improved in Gantai-treated group as compared with the model group. Collagen fibers of the liver were markedly decreased in Gantai-treated group as compared with the model group. The Gantai-treated group had lower levels of serum hyaluronic acid, ALT, T-CHOL and γ-GT than the model group. Liver function was significantly improved in Gantai-treated group. Conclusion Gantai capsules are effective in the treatment of alcohol-induced liver fibrosis by attenuating hepatic injury and fibrosis.
