摘要
目的:研究黄芪葛根汤对实验性糖尿病心肌病变的干预作用及机制。方法:采用高脂饲料喂养和链脲佐菌素腹腔注射的方法建立2型糖尿病大鼠心肌病变模型,观察黄芪葛根汤对血糖、胰岛素、心肌纤维化、转化生长因子-β1(TGF-β1)及其信号蛋白Smad3表达的影响;mRNA表达采用RT-PCR方法检测,蛋白表达采用免疫组化方法。结果:黄芪葛根汤可显著降低糖尿病大鼠空腹血糖、胰岛素,改善胰岛素抵抗,减少心肌羟脯氨酸含量抑制胶原增生,降低心脏指数,并显著抑制心肌TGF-β1/Smad3 mRNA和蛋白的强表达。结论:黄芪葛根汤可以通过调控TGF-β1/Smad3通路显著干预实验性糖尿病心肌纤维化病变。
Objective:To study the effect of HGD on diabetic cardiomyopathy and its mechanism. Methods:The T2-DM rats model was established by combining high fat diet with STZ. The blood glucose, insulin, myocardial fibrosis and TGF-β1/Smad3 signaling path- way were observed; TGF-β1 and Smad3 mRNA expression were detected by RT-PCR method, protein expression detected by immunohis- tochemical method. Results: HGD obviously reduced fasting blood glucose, insulin, improved insulin resistance, reduced myocardial hydroxyproline contents, lowered cardiac index, significantly inhibited over-expression of TGF-β1/SMAD3 mRNA and protein in diabet- ic rats cardiac. Conclusion:HGD can obviously prevent experimental diabetic myocardial fibrosis through the regulation effect on TGF-β1/Smad3 signaling pathway.
