摘要
目的:观察野黄芩苷对人舌鳞癌Tca8113细胞周期及Fas蛋白表达的影响。方法:用浓度为80、120和160μg/ml的野黄芩苷处理体外培养的人舌鳞癌Tca8113细胞,用流式细胞技术观察细胞周期的变化;用蛋白印迹法定量检测Fas蛋白表达。结果:与对照组相比,加入不同浓度野黄芩苷后DNA合成前期(G1)细胞所占百分比(G1%)升高(P<0.05),160μg/ml野黄芩苷处理后DNA合成期(S)细胞所占百分比(S%)降低(P<0.05)。蛋白印迹法结果显示,随着药物浓度的增加,Fas蛋白表达量有增高趋势,与对照组比较,差异有统计学意义(P<0.05)。结论:野黄芩苷可能通过抑制肿瘤细胞增殖,提高人舌鳞癌Tca8113细胞Fas蛋白的表达而达到抗肿瘤的作用。
Objective: To investigate the effects of scutellarin on the Fas expression and cell cycle distribution of human tongue squa- mous cell carcinoma Tea-8113 cells. Methods: Tca-8113 cells were cultured in vitro and treated by scutellarin at various concentra- tions. Flow cytometry was used to examine the cell cycle distribution. Western blot was used to detect the Fas expression of the cells. Results: Scutellarin at 80, 120 and 160 μg/ml increased the percentage of G1-phase cells (P 〈0. 05 ) and at 160 μg/ml decreased the percentage of S- phase cells ( P 〈 0.05 ). Scutellarin at 80, 120 and 160μg/ml increased the Fas expressions ( P 〈 0. 05 ). Conclusion: Scutellarin may inhibit the proliferation and enhance the Fas expressions of human tongue squamous cell carcinoma Tea-8113 cells.
