摘要
目的:研究丹皮酚对氧应激致人源肝星状细胞(Hepatic stellate cell,HSC)LX2增殖及TGF-β1、Smad4和Collagen I mR-NA表达的影响。方法:在建立体外过氧化氢(H2O2)致HSC-LX2细胞氧应激模型基础上,用MTT法检测丹皮酚对HSC-LX2细胞增殖的影响,生化检测HSC-LX2细胞内MDA及SOD,荧光定量PCR检测丹皮酚对氧应激致HSC-LX2细胞TGF-β1、Smad4和Collagen ImRNA表达的影响。结果:丹皮酚呈剂量依赖性抑制H2O2诱导HSC-LX2增殖,抑制细胞MDA含量及增加SOD活性,以50μmol/L浓度时最为明显,与DMSO组比较差异有显著性(P<0.01);丹皮酚各剂量组作用24 h后,HSC-LX2细胞中TGF-β1、Smad4和Collagen ImRNA的水平降低,与DMSO组比较(P<0.01),且作用呈剂量依赖性。结论:丹皮酚预处理肝星状细胞可下调TGF-β1、Smad4mRNA的表达,进而抑制肝星状细胞Collagen I的合成,发挥抗肝纤维化作用。
Aim: To investigate the effect of paeonol on the expression of TGF-β1, Smad4 and Collagen I mRNAinhuman he- patic stellate cells ( HSC - LX2) activated by H2 O2. Methods: HSC - HSC - LX2 were cultured in vitro, stimulated by H2 O2 for 2 hours and treated with different concentrations of paeonol for 24 hours, MTY assay was used to evaluate the inhibitive effect of paeonol. The expressions of TGF - β1, Smad4 and Collagen I mRNA were determined by Real - time quantitative PCR. Results Different con- centration of paeonol inhibited the activation and proliferation of HSC - LX2, in a dose dependent manner. The inhibitive effect was most obvious in 50 μmol/L. he expressions of TGF -β1, Smad4 and Collagen I were markedly reduced by the treatment with three concentrations of plumbago than that in the leptin group, and was dose dependent. Conclusion The expressions of TGF - β1, Smad4 can be down - regulated by paeonol, resulting in the reduced synthesis of Collagen I, which may be one of the possible molecular mechanism against hepatic fibrosis.
