丙戊酸钠联合紫杉醇对宫颈癌Hela细胞抑制作用的研究 被引量:2

摘要 目的探讨组蛋白去乙酰化酶抑制剂丙戊酸钠(valproic acid,VPA)联合紫杉醇对宫颈癌Hela细胞的体外抑制作用。方法 VPA和紫杉醇单独及联合作用于宫颈癌Hela细胞,显微镜下观察细胞形态变化,采用四甲基偶氮唑蓝法(MTT)检测细胞增殖,应用流式细胞术检测细胞凋亡率及细胞周期变化。结果 VPA对宫颈癌Hela细胞株有明显生长抑制作用,呈时间剂量依赖性,使细胞凋亡增强,细胞周期阻滞于G2/M期;VPA联合紫杉醇后,宫颈癌Hela细胞凋亡率明显增高,细胞周期进一步阻滞于G2/M期,S期细胞数减少。结论 VPA可显著抑制人宫颈癌Hela细胞的增殖并诱导细胞凋亡,增强紫杉醇对Hela细胞的化疗作用。 Objective To explore the inhibitory effect of histone deacetylase inhibitor VPA combined with paclitaxel on human cervical cancer Hela cells. Methods Hela cells were treated with VPA and paclitaxel alone and in combination. The cell condition was observed by microscope. Cell proliferation was studied by MTT assay. The apoptosis and cell cycle were determined by flow cytometry. Results The Hela ceils were significantly inhibited by VPA in a time and dose-dependent manner,and VPA induced enhanced apoptosis and G2/M phase arrest. When combined with paclitaxel, VPA induced more severe apoptosis and ar- rested much more cells at G2/M phase;meanwhile the cells of S phase were decreased. Conclusion VPA can significantly inhib- it the proliferation of Hela cells and induce apoptosis, enhancing paclitaxel's killing effect on Hela cells.
