肝纤维化进程中miRNA-19b表达变化的研究 被引量:3

Expression of miRNA-19b in Hepatic Fibrosis Rats

摘要 目的:观察微小RNA(miRNA)-19b在肝纤维化大鼠肝脏组织的表达,探讨miRNA-19b在肝纤维化进程中的作用。方法:将20只SD大鼠随机分成正常对照组和肝纤维化模型组。采用改良式复合因素法复制大鼠肝纤维化模型,分别在6、7周后,取各组大鼠肝脏组织,用荧光实时定量PCR方法检测miRNA-19b的表达。结果:6周后肝纤维化模型组大鼠肝脏组织miRNA-19b的表达低于正常对照大鼠组(P<0.05),7周模型组大鼠肝脏组织miRNA-19b的表达低于6周模型组(P<0.05)。结论:miRNA-19b在肝纤维化大鼠肝脏组织中表达降低,可能与纤维化进程相关,且可能成为肝纤维的诊断以及预后的一个指标。 Objective: To study the effects of miRNA-19b through observing the expression of miRNA-19b in the liver on hepatic fibrosis(HF) rats.Methods: 20 rats were randomly divided into the normal group,HF group.The HF model in rats was duplicated with Improved Composite Factor Method.The liver tissues were taken after 6 weeks and 8 weeks.MiRNA-19b was tested by florescent real-time quantitative PCR.Results: Compared to the normal control rats,the level of miRNA-19b was markedly decreased in HF rats after 6 weeks(P0.05).besides that,the level of miRNA-19b was more prominently decreased in HF rats after 7 weeks than 6 weeks(P0.05).Conclusions: MiRNA-19b was decreased in liver tissues of hepatic fibrosis rats.It probably involved in the progress of HF and was the marker of diagnosis and prognosis of HF.
