摘要
为了克隆被孢霉不饱和脂肪酸生物合成途径的相关酶基因 ,构建了基于λgt1 0的被孢霉cDNA文库。cDNA文库库容量为 2× 1 0 6pfu。以已克隆的被孢霉△9脂肪酸脱饱和酶保守区cDNA为探针对被孢霉cDNA文库进行筛选。经过两轮筛选获得 1个阳性克隆 ,其插入片段长度大于 1 6kb。
Mortierella\% species have potential for fermentative production of polyunsaturated fatty acids including \|linolenic acid, Arachidonic acid and EPA, etc. In order to clone genes encoding enzymes in the unsaturated fatty acid biosynthetic pathway, cDNA library of Mortierella was constructed using λgt10 vector. Using cDNA encoding conserved region of △ 9 fatty acid desaturase gene as probe,\%Mortierella\% cDNA library was screened. After two rounds of screening one positive clone was identified which has insert length of larger than 1.6kb.
