摘要
目的:探讨脑胶质瘤中Survivin和Fhit的表达及与MRI瘤周水肿的关系。方法:将70例术前无放化疗史的脑胶质瘤手术标本和10例正常脑组织,利用采用免疫组化法分别检测Survivin和Fhit的表达,MRI瘤周水肿程度的影像学分级依据中华医学会神经外科分会2009年专家共识征求意见稿。结果:在脑胶质瘤中Survivin蛋白阳性率为64.28%,明显高于对照组(0.0%),其表达率随着肿瘤级别的增高表达增强。Fhit蛋白的阳性率为51.42%,明显低于对照组(100%),其表达率随着肿瘤级别的增高而递减。Survivin蛋白阳性率与MRI瘤周水肿呈正相关。Fhit蛋白阳性率与MRI瘤周水肿呈负相关。结论:Survivin和Fhit均参与细胞周期调控和细胞凋亡,两者的生物学效应相反,从而影响胶质瘤的发生发展。对二者进行联合检测可为进一步研究胶质瘤提供必要的理论依据。同时在术前根据MRI瘤周水肿的影像学资料可以对肿瘤的分级进行初步评估。
Objective:To explore expression of Survivin and Fhit, and their relationship with peritumoral edema on MRI in glioma. Methods:The sample were composed of tissue form 70 cases of glioma carcinoma without a history of chemo-radiation therapy and 10 cases of normal brain tissue of control subject. Immunohistochemistry were employed to determine the expression of Survivin and Fhit. The level of peritumoral edema on MRI in glioma is base on the literature of neurosurgery branch of Chinese Medical Associaition in 2009. Results:The positive rate of Survivin in glimoa was 64.28%, and it was much higher than control tissue. The protein expression of Survivin in glioma was positive related with malignancy of glioma. The positive rate of Fhit in glioma was 51.42% ,it was much lower than control tissue. The protein expression of Fhit in glioma was decreased in high-grade glioma. The expression of survivin was positive relative with peritumoral edema on MRI in glioma. The expression of Fhit was negatively relative with peritumoral edema on MRI in glioma. Conclusion: Survivin and Fhitall take part in the regulation of the biologic process of glioma cells. Their biological effect was opposite, combined detection of Surviving and Fhit may offer a necessary theory base for study of glioma further. Analyze the information of peritumoral edema on MRI preoperative could have a evaluation of glioma grading.
