摘要
目的采用高效液相色谱法建立复方硫酸双肼屈嗪三组分的含量测定方法。方法采用Phe-nomenC18柱(250×4.6 mm,5μm),流动相0.2%辛烷磺酸钠溶液(用磷酸调节pH值至3.5);甲醇∶乙腈(75∶23∶2),检测波长220 nm,流速1.0 ml/min,柱温30℃,采用外标法计算。结果硫酸双肼屈嗪的线形关系为57.6~460.8μg/ml(r=0.999 9),平均回收率为99.7%,RSD为1.1%(n=9);氢氯噻嗪的线形关系为50.0~400.0μg/ml(r=0.999 7),平均回收率为97.8%,RSD为0.9%(n=9);盐酸可乐定的线形关系为0.625~1.0μg/ml(r=0.999 5),平均回收率为98.3%,RSD为0.7%(n=9)。结论 HPLC法同时测定硫酸双肼屈嗪、氢氯噻嗪、盐酸可乐定的含量,方法专属性好,灵敏度高,结果可靠,对进一步完善现有质量标准具有借鉴价值。
Objective To establish the compound sulfate hydralazine three - component determination by using high perform-ance liquid chromatography (HPLC). Methods PhenomenC18 column (250 × 4.6 mm, 5μm) was used for determination of the con-tent of sulfuric acid double hydralazine three components; Mobile phase: 0.2% octane sulfonate solution (adjusted with phosphoric acid to pH 3.5 ) ; methanol : acetonitrile (75:23:2) ; detection wavelength : 220nm; flow rate : 1.0 ml/min. Column temperature of 30℃, using the external standard method. Results The linear relationship of the sulfuric acid double hydrazine flexor triazine was 57.6 - 460.81μg/ml (r = 0.999 9), the average recovery rate was 99.7%, RSD was 1.1% (n =9) ; hydrochlorothiazide thiophene triazine of the linear relationship was between 50.0 to 400.0 μg/ml ( r = 0. 999 7 ), and the average recovery was 97.8%, RSD was 0.9% ( n = 9) ; the clonidine hydrochloride linear relationship was 0.625 to 1.0 μg/ml (r =0.999 5), the average recovery rate was 98.3% and the relative standard deviation was O. 7% ( n = 9 ). Conclusion This study shows that HPLC method can simultaneously determine the content of sulfate hydralazine, hydrochlorothiazide and clonidine hydrochloride, and this method is specificity, high sensitivity and relia-ble, with a reference value to further improve the existing quality standards.
