丹参注射液对急性脑梗死大鼠脑组织VEGF表达的影响 被引量:16

Effects of Danshen Injection on the Expression of Vascular endothelial Growth Factor in rats with Acute Cerebral Infarction

摘要 目的观察丹参注射液对急性脑梗死大鼠脑组织中血管内皮生长因子(VEGF)表达的影响。方法用颈内动脉线栓法制备大脑中动脉闭塞大鼠模型,入选大鼠随机分为3组:丹参组、对照组及假手术组。丹参组:于造模成功至实验取材期间每日腹腔注射丹参注射液(0.5mL/次),连用7d,采用双抗体夹心酶联免疫法(ELISA)检测各组中不同时段(3d、7d)脑组织VEGF的水平,并进行神经功能缺损评分(NSS);对照组:使用生理盐水;假手术组:不用药。结果丹参组不同时段脑组织VEGF水平高于对照组、NSS评分低于对照组(P<0.05)。结论丹参注射液能够上调脑梗死大鼠脑组织VEGF的表达及改善神经功能,对于加强血管发生及神经重塑的联系有积极作用。 Objective To explore the effects of Danshen injection(DSI) on the expression of cerebral vascular endothelial growth factor(VEGF) in rats with acute cerebral infarction(ACI). Methods The middle cerebral artery occlusion (MCAO) model was estab- lished by the method of Zea Longa. Sixty rats were randomly divided into 3 groups : DSI group, the control group and the sham - operated group. The rats in DSI group were injected intraperitoneally with DSI(0. 5 mL,once a day) for 7 days from the success of the modeling to experimental drawn. Cerebral VEGF concerntrations were measured by double antibody sandwich ELISA on day 3,7 after the onset of ACI, Neurological Severity Score were performed at 1,3 and 7days after MCAO. The rats in control group were injected intraperitoneally with saline. The rats in the sham - operated group without any drug. Results The content of cerebral VEGF in DSI group was significantly higher than that in the control group, NSS scores were lower than that in the control group(P〈0.05). Conclusion DSI could promote cerebral VEGF expression in MCAO rats and improve nerve function. There was positive impact to strengthen the contact between angiogenesis and neural remodeling,which was worth to expected to improve the therapeutic effect.
