摘要
目的探讨黄芪丹参"药对"对大鼠肾间质纤维化的保护作用及其干预肾纤维化抑制因子par-3的量效关系。方法将40只雄性SD大鼠随机分为正常组(n=8)和造模组(n=32)。造模组采用单侧输尿管梗阻(UUO)建立肾间质纤维化模型,造模后分为模型组(n=8),黄芪丹参低剂量组(n=8)、中剂量组(n=8)、高剂量组(n=8)。造模后第2天开始各组给予相应的干预措施;2周后处死大鼠,收集相应标本,检测肾功能、尿α1-微球蛋白,观察肾组织病理改变,并应用Western Blot方法检测肾组织par-3蛋白的表达情况。结果①与正常组比较,模型组及各治疗组血清Cr明显高于正常组(P<0.05);除高剂量组BUN水平无差异外,其余各组血清BUN明显高于正常组(P<0.05)。②与模型组比较,各治疗组尿α1-微球蛋白均降低(P<0.05),各治疗组间差异无统计学意义(P>0.05)。③与模型组比较,各治疗组肾脏组织病理情况均有不同程度的改善。④黄芪丹参"药对"可以促进par-3的表达,并且随用药剂量的增高表达量增加。结论黄芪丹参"药对"可通过促进par-3的表达,从而保护肾小管功能,减轻肾间质纤维化的进展,并呈一定的量效关系。
Objective To observe the protective effect of the paired-herb of Radix Astragali seu Hedysarion and Radix Salviae Miltiorrhizae on par-3, and explore its action and dose-effect relationship. Methods Forty male SD rats were randomly divided into normal group ( n = 8 ) and model group ( n -- 32 ). The rats in model group were established by unilateral ureteral obstruction (UUO) and divided into the control group (n = 8 ), small-dose paired group (n =8 ), medium-dose group ( n = 8 ), large-dose group ( n = 8 ). Two days after establishment of the model, corresponding interventions were given to each group, and then the rats were killed for samples collection. The renal function, urinary α1-microglobulin, renal pathological changes were observed, and the expression of par-3 in rental tissue was detected by Western blotting. Results ① The serum Cr value in model group and treatment groups were markedly higher than that of the normal group ( P 〈 0.05 ) ; except the high-dose group, the serum BUN was significantly higher in all model groups than normal group ( P 〈 0.05 ) ;② Compared with the model group, the concentration of cd-microglobulin in treatment group was decreased ( P 〈 0.05 ), no significant difference among all treatment groups ( P 〉 0.05 ). ③Compared with control group, the pathological changes of renal tissue were improved in all treatment groups. ④ The paired-herb of Radix Astragali seu Hedysarion and Radix Salviae Miltiorrhizae can promote the expression of par-3, and the expression increased with the increase of the dose. Conclusion The paired-herb of Radix Astragali seu Hedysarion and Radix Salviae Miltiorrhizae can protect the renal tubular function and reduce the development of renal fibrosis by promoting expression of par-3 in a dose dependent manner.
